枯草芽孢杆菌发酵中药皂素共表达蛋白酶及果胶酶的研究

Study on Protease and Pectinase Co-expressed by Bacillus subtilis in Fermentation of Chinese Medicine Saponin

  • 摘要: 为促进中药皂素的开发利用,本文采用单因素方法设置不同提取时间、温度、pH、料液比,以富含皂苷的4种中草药的混合水提液作为发酵基质用共表达蛋白酶和果胶酶的枯草芽孢杆菌或添加5%葡萄糖进行发酵,测定发酵前后发酵基质的洗涤效果、透光率和皂素变化。结果表明:提取中药皂素的条件为60℃,3h,pH9.0,料液比1∶18。与发酵前相比,经枯草芽孢杆菌直接发酵后的皂素提取液洗涤效果提升了8.18%。添加5%葡萄糖后得到的发酵液的洗涤效果与蛋白酶及果胶酶的酶活呈正相关。两种发酵方式得到的发酵液透光率均在发酵32h达到最大。发酵前后皂素基本稳定不变。研究发现中药皂素经枯草芽孢杆菌发酵后的酵素液可以作为洗涤液的前期开发。

     

    Abstract: In order to promote the development and utilization of traditional Chinese medicine(TCM)saponin,this article using four kinds of Chinese herbal medicine that are rich in saponins of mixed water extraction liquid as fermentation substrate with a total expression of bacillus subtilis protease and pectinase or add 5% glucose to fermentation.The mixed water extraction liquid was tested by single factor method to set different extracting time,temperature,pH,and solid-liquid ratio.The determination of the washing effect of fermentation before and after fermentation substrates,light transmittance and saponin was tested then.The results showed that he extraction conditions of traditional Chinese medicine saponin were 60℃,3 h,pH9.0 and the ratio of solid to liquid 1∶18.The washing effect of the saponin extract directly fermented by bacillus subtilis increased by 8.18%compared with that before fermentation.The washing effect of fermentation broth obtained by adding 5% glucose was positively correlated with the activity of protease and pectinase.The light transmittance of the fermentation broth obtained by the two fermentation methods reached the maximum at 32 h.The saponin was basically stable before and after fermentation.In conclusion,the fermentation liquid of traditional Chinese medicine saponin fermented by bacillus subtilis can be used as the early development of washing liquid.
