应用正交试验建立树莓组织培养快繁体系

Establishment of Rapid Propagation System in Raspberry Tissue Culture by Orthogonal Experiment

  • 摘要: 为促进树莓工厂化育苗,本试验采用俄罗斯引进的树莓品种"双季2381"和"单季1132"为材料,以茎段为外植体,应用正交试验对最佳腋芽萌发培养、丛生芽增殖及组培苗生根培养基进行筛选,建立树莓快繁技术体系。结果表明:腋芽萌发最佳培养基"双季2381"为6-BA 1mg·L-1+NAA 0.3mg·L-1,"单季1132"为6-BA 1.0mg·L-1+TDZ 0.5mg·L-1+NAA 0.5mg·L-1,其萌芽率分别为91.7%和92.3%,且腋芽萌发快、叶色绿。丛生芽增殖最佳培养基:"双季2381"为6-BA 2.0mg·L-1+TDZ 0.5mg·L-1+NAA 0.1mg·L-1,"单季1132"为6-BA 1.0mg·L-1+TDZ 1.0mg·L-1+NAA 0.3mg·L-1,其增殖系数分别为6.7和6.2,且丛生芽增殖快,叶色绿。组培苗生根最佳培养基"双季2381"和"单季1132"均为1/2MS+IBA 0.2mg·L-1,其生根率分别达96.9%和95.1%,且自培苗生根快,叶色绿。

     

    Abstract: To promote the factory seedling cultivation of raspberry,the raspberry varieties of"Double-season2381"and"Single-season 1132",introduced from Russia were used as experimental materials.The stem segment was used as explants.The orthogonal experiment was used to screen the best culture medium of axillary bud germination,cluster bud proliferation and tissue culture seedling rooting,so as to establish the technology system of rapid propagation.The results showed that the best medium of axillary bud germination"Double-season 2381"was 6-BA 1 mg·L-1+NAA 0.3 mg·L-1,and "Single-season 1132"was 6-BA 1.0 mg·L-1+TDZ0.5 mg·L-1+NAA 0.5 mg·L-1,the germination rate was 91.7% and 92.3%,respectively,the growth rate of the axillary buds was fast and the leaf color was green.The best medium of cluster bud proliferation"Doubleseason 2381"was 6-BA 2.0 mg·L-1+TDZ 0.5 mg·L-1+NAA 0.1 mg·L-1,"Single-season 1132"was 6-BA1.0 mg·L-1+TDZ 1.0 mg·L-1+NAA 0.3 mg·L-1,the multiplication coefficients were 6.7 and 6.2,respectively,the growth rate of cluster buds was fast and the leaf color was green.The best medium of tissue culture seedling rooting"Double-season 2381"and "Single-season 1132"were 1/2 MS+IBA 0.2 mg·L-1,the rooting rate was 96.9%and 95.1%,respectively,the rooting speed of tissue culture seedlings was fast and the leaf color was green.
