玉米鞘腐病菌ISSR-PCR反应体系优化

Optimization of ISSR-PCR Reaction System of Fusarium proliferatum

  • 摘要: 玉米鞘腐病的主要病原为层出镰孢(Fusarium proliferatum),为明确其ISSR-PCR最佳反应体系,本文以层出镰孢DNA为模板,筛选各因子浓度范围,利用单因素试验,对5因素进行优化,确定最佳反应体系,从13条供试引物中筛选扩增条带清晰、多态性好的引物,并筛选最适退火温度。结果表明:ISSR-PCR最佳反应体系各因子浓度分别为模板DNA 40ng·μL-1、镁离子浓度2.5mmol·L-1、引物浓度为0.4μmol·L-1、dNTPs浓度0.2mmol·L-1、Taq酶浓度为2.5U·μL-1,最适ISSR引物为808,最适退火温度是58℃,用筛选的最适引物808对供试菌株扩增出14条条带,多态性条带13条,多态性条带比率为92.90%。

     

    Abstract: The major pathogen of corn sheaths rot is Fusarium proliferatum.In order to clarify the optional response system of ISSR-PCR of F.proliferatum.DNA was used as a template to screen the concentration range of each factor,and orthogonal test was used to optimize 5 factors to determine the best response system.The primers with clear amplified bands and good polymorphism were selected from 13 primers and selected optional annealing temperature.The results showed that ISSR-PCR optional reaction system:template 40 ng·μg-1,magnesiumion concentration 2.5 mmol·L-1,primer concentration 0.4μmol·L-1,dNTPs concentration0.2 mmol·L-1,Taqenzyme concentration 2.5 U·μL-1,the optional primer was 808 and optional annealing temperature was 58℃,a total of 14 bands were amplified with the screening out of primers 808.And polymorphic bands were 13 among them.The percentage of polymorphic bands was 92.9%.
