Abstract:
In order to optimize the disinfection system in tissue culture of wild Platycodon grandiflorum(Jacq.) A. DC. from Jilin Xianghai National Nature Reserve, NaClO disinfection method was used to determine the sterilization concentrations, sterilization time and culturing temperature of getting aseptic seedlings; the methods of hormone-seed soaking and heat-shock treatment were used to select the factors of hormone types, soaking concentrations as well as treating temperature and time, which affect the conditions for seed germination of wild Platycodon grandiflorum(Jacq.) A. DC.. Results showed that the optimum conditon to get aseptic seedling system of wild Platycodon grandiflorum(Jacq.) A. DC.was composed of soaking seeds by sterile wate, heat shocking treatment at 55 ℃ for 10 min, disinfected treatment in NaClO(1%) for 20 min and cultivated under visible light condition at 15 ℃, by which the regeneration rate of aseptic seedlings was 90.00%. The resnlts indicated that high levels seed germinating percentage and high quality aseptic seedlings of wild Platycodon grandiflorum(Jacq.) A. DC. could be got by means of dormancy-release method with disinfection treatments.
