Abstract:
In order to study the effect of different conditions on the sporulation process of Toxoplasma gondii oocysts, the experiment was conducted to feed the cats with fresh pig liver, and to separate the oocysts of Toxoplasma gondii from the feces; under aerobic and anaerobic conditions, it was incubated at different temperatures 4 ℃,room temperature(18~22 ℃),25 ℃ and 37 ℃, sulfuric acid solution concentration(1%,2%,4%) and pH values(1-14);on the 2 nd, 4 th, 6 th, 8 th, 10 th, 12 th, 14 th, 16 th, 18 th, 20 th, 22 th, and 24 th day of culture, the sporulation of Toxoplasma gondii oocysts was observed under microscopy. The results showed that the Toxoplasma gondii oocysts were successfully isolated on the 6 th day of feeding the cat. Under aerobic conditions, Toxoplasma gondii oocysts were cultured at 4 ℃ until it did not sporulate on the 24 th day; at room temperature, with the pH value of 4,5,10,12,13,and 14,it began to sporulate on the second day; at 25 ℃ and the pH value of 4,it began to sporulate on the second day; at 37 ℃ and the pH value of 1,2,3,4,9,12,13,it began to sporulate on the second day. Under anaerobic conditions, at 4 ℃ and the pH values of 1 and 3,Toxoplasma gondii oocysts began to sporulate on the 12 th day; at room temperature, the concentration of sulfuric acid solution of 1%,2%,and the pH value of 1,2,3,4,5,11,12,13,14,it began to sporulate on the 14 th day; at 25 ℃,sulfuric acid solution concentration of 1%,4%,and the pH value of 1,it began to sporulate on the 12 th day; at 37 ℃,it was not sporulated on the 24 th day. The results suggested that 25 ℃ was the optimum temperature for the oocyst sporulation of Toxoplasma gondii;4 ℃ can inhibit the oocyst sporulation of Toxoplasma gondii;oxygen was beneficial to the oocyst sporulation of Toxoplasma gondii;pH value and sulfuric acid solution concentration had low effect on the oocyst sporulation of Toxoplasma gondii.
