平原型和田羊毛囊KAP3.2基因的克隆与序列分析

Cloning and sequence analysis of hair follicle KAP3.2 gene of plain Hetian sheep

  • 摘要: 为了研究平原型和田羊毛囊角蛋白关联蛋白(keratin associated protein, KAP)3.2基因对半粗毛羊羊毛经济性状的影响,试验以新疆南疆地方品种平原型和田羊皮肤毛囊为材料,提取皮肤毛囊总RNA,反转录得到cDNA并进行PCR扩增,构建重组克隆质粒pMD18-T-KAP3.2和重组表达质粒pET-28a-KAP3.2,并运用生物信息学软件对该基因进行分析。结果表明:试验成功克隆出平原型和田羊毛囊KAP3.2基因,该基因片段长度为306 bp,编码98个氨基酸,其与GenBank中美利奴羊(登录号为M21099.1)核苷酸序列的同源性为98.7%,有4处碱基发生突变,分别是第104位的G突变为C、第138位的C突变为T、第226位的C突变为G、第265位的C突变为G。通过翻译对比后平原型和田羊KAP3.2基因编码的氨基酸序列有3处氨基酸发生突变,分别是第35位的丝氨酸突变为苏氨酸,第76,89位的精氨酸突变为甘氨酸。说明平原型和田羊毛囊KAP3.2基因序列虽有个别碱基发生了变化,但仍高度保守,与GenBank中美利奴羊的同源性较高,亲缘关系较近。

     

    Abstract: In order to study the influence of keratin associated protein(KAP) 3.2 gene on the economic characters of semi-coarse-wool sheep wool, the total RNA was extracted from skin hair follicles of plain Hetian sheep, a local breed in southern Xinjiang Province, and cDNA was obtained by reverse transcription and PCR amplification was performed. The recombinant clone plasmid pMD18-T-KAP3.2 and the recombinant expression plasmid pET-28 a-KAP3.2 were constructed, and bioinformatics software was used to analyze the gene. The results showed that KAP3.2 gene of plain Hetian sheep hair follicle was successfully cloned, the gene fragment length was 306 bp, encoding 98 amino acids. The sequence of this gene had 98.7% homology with the nucleotide sequence of Merino sheep(M21099.1) in GenBank. There were four mutations, namely, the mutation of G at position 104 was C, the mutation of C at position 138 was T, the mutation of C at position 226 was G, and the mutation of C at position 265 was G. Through translation comparison, there were three mutations in the amino acid sequence encoded by the KAP3.2 gene of plain Hetian sheep. The amino acid sequence was mutated from serine to threonine at position 35, from arginine to glycine at position 76 and 89.The results indicated that KAP3.2 gene sequence of plain Hetian sheep hair follicle was highly conserved although some bases were changed. KAP3.2 gene sequence was homologous and closely related to Merino sheep in GenBank.
