氨气对4~6周龄肉鸡免疫器官中MUC5AC及E-cd基因表达的影响

The effect of ammonia on the expression of MUC5AC and E-cd genes in immune organs of 4-6 weeks old broilers

  • 摘要: 为了研究鸡处于高浓度氨气环境中免疫系统的变化,试验采用实时荧光定量PCR方法检测经15,30,60,0 mg/m3氨气处理(A~D组)后的4~6周龄肉鸡法氏囊、胸腺及脾脏等免疫器官中相关免疫基因MUC5AC和E-cd的表达情况。结果表明:6周龄时,B组肉鸡法氏囊中MUC5AC与E-cd基因的相对表达水平为D组的10倍左右(P<0.05);胸腺中MUC5AC基因相对表达水平为D组的5倍左右(P<0.05),E-cd基因相对表达水平也升高但与D组比较差异不显著(P>0.05);脾脏中MUC5AC基因相对表达水平也升高但与D组比较差异不显著(P>0.05),而E-cd基因相对表达水平与D组比较差异显著(P<0.05)。B组4周龄肉鸡法氏囊中E-cd和MUC5AC基因相对表达水平升高,6周龄肉鸡胸腺中E-cd和MUC5AC基因相对表达水平也有升高趋势;C组各周龄肉鸡脾脏中MUC5AC基因的相对表达水平降低。说明在肉鸡长时间受到氨气刺激时,MUC5AC和E-cd基因的表达水平存在器官差异,法氏囊具有囊腔,与胸腺和脾脏相比,E-cd基因的总体相对表达水平升高,这与上皮细胞结构的完整性有关。

     

    Abstract: In order to study the changes in the immune system of chickens in a high-concentration ammonia environment, the experiment used real-time fluorescence quantitative PCR to detect the relevant immune gene expression(MUC5 AC and E-cd genes) in the immune organs(bursa of Fabricius, thymus and spleen) of broilers treated with ammonia(15,30,60,0 mg/m~3) the situation. The results showed that at 6 weeks of age, compared with the D group, the relative expression levels of MUC5 AC and E-cd genes in the bursa of Fabricius of broilers in the B group increased by about 10 times(P<0.05);and the relative expression level of MUC5 AC gene in thymus increased by about 5 times(P<0.05) the relative expression level of E-cd gene in the thymus also increased but with no significant difference compared with D group(P>0.05),and the relative expression level of MUC5 AC gene in the spleen also increased but with no significant difference compared with D group(P>0.05),and the relative expression level of E-cd gene in the spleen with significant difference compared with D group(P<0.05). The relative expression levels of E-cd and MUC5 AC genes was increased in bursa of Fabricius of broilers at 4 weeks of age in the B group, and the relative expression levels of E-cd and MUC5 AC genes was increased in the thymus of broilers at 6 weeks of age in the B group; The relative expression level of MUC5 AC gene in the spleen of broilers with each age was decreased in the C group. The results suggested that there were organ differences in the expression levels of MUC5 AC gene and E-cd gene of broilers were stimulated with ammonia for a long time. The bursa of Fabricius had a cavity, and the overall relative expression level of E-cd gene was elevated compared with thymvs and spleen, which was related to the structural integrity of epithelial cells.
