Abstract:
In order to screen for compound immune adjuvants that could enhance the immune effect of subunit vaccines, the ZJ01 and ZJ02 adjuvants screened in the previous study were used and were mixed with immunopotentiators A,B,C,and D respectively to prepare compound adjuvants. The compound adjuvants were then mixed with a subunit vaccine of ZIKV E protein for Balb/c mice immunization to detect the levels of IgG,IgG1,IgG2 a, and IgG2 b antibodies, the levels of ZIKV E protein specific antibodies and neutralizing antibodies, and the effect of lymphocyte proliferation in mice after immunization, and to verify the immune enhancement effect of the compound adjuvants. The results showed that on the 35 th day after immunization, the serum levels of IgG,IgG1,IgG2 a, IgG2 b antibodies, specific antibodies and neutralizing antibodies in the ZJ01 and ZJ02 test groups containing components A and D were mostly significantly or extremely significantly higher than E Protein control group(P<0.05 or P<0.01,P<0.001,P<0.000 1),in which IgG,IgG1,IgG2 a, and IgG2 b antibody levels were 1.74-3.35 times than those of the E protein control group; the specific antibody level was 1.43-1.60 times than that of E protein control group; the neutralizing antibody level was 1.33-1.75 times than that of the E protein control group; the T lymphocyte proliferation level was 1.30-1.36 times than that of the E protein control group. The results indicated that the compound adjuvants of ZJ01 and ZJ02 containing A and D components could better enhance the immune effect of subunit vaccine.
