Abstract:
To investigate the effects of ellagic acid on apoptosis of porcine intestinal epithelial cells(IPEC-J2) and the expression levels of related genes Bcl-2, Bax, caspase-3, caspase-8 and caspase-9, IPEC-J2 was treated with 20, 40, 60, 80 μg/mL ellagic acid for 24 hours, and 0 was added as control. The effects of different concentrations of ellagic acid on cell proliferation were detected by the method of tetramethylazazolate(MTT), the rate of cell apoptosis was determined by flow cytometry, the content of caspase-3 was determined by ELISA, and the mRNA expression of Bcl-2, Bax, caspase-8 and caspase-9 were determined by on intestinal epithelial cells real-time fluorescence quantitative PCR(qRT-PCR). Results show that the ellagic acid plays a protective role on intestinal epithelial cells, compared with 0 to add, different concentration of ellagic acid cells after 24 h, cell apoptosis rate, the expression levels of Bcl-2, Bax, caspase 3, caspase-8, and caspase-9 were significantly or extremely significantly decreased(P<0.05 or P<0.01). It indicated that the ellagic acid resistant mechanism of apoptosis and inhibition had related to the expression levels of Bax, Bcl-2, caspase 3, caspase-8, the expression of caspase-9.
