鞣花酸对IPEC-J2细胞凋亡及相关基因表达的影响

Effects of ellagic acid on apoptosis and apoptosis-related gene expression in IPEC-J2 cells

  • 摘要: 为了探究鞣花酸对猪肠道上皮细胞(IPEC-J2)凋亡和相关基因Bcl-2、Bax、caspase-3、caspase-8、caspase-9表达的影响,试验用浓度为20,40,60,80μg/mL的鞣花酸处理IPEC-J2 24小时,并以不添加作为对照,采用四甲基偶氮唑盐(MTT)法检测不同浓度鞣花酸对IPEC-J2增殖的影响,用流式细胞术测定IPEC-J2凋亡率,用ELISA方法检测caspase-3的含量,用实时荧光定量PCR(qRT-PCR)方法检测Bcl-2、Bax、caspase-8、caspase-9 mRNA的表达情况。结果表明:鞣花酸对IPEC-J2具有保护作用,与不添加相比,不同浓度鞣花酸作用IPEC-J2 24 h后,IPEC-J2凋亡率、Bcl-2、Bax、caspase-3、caspase-8、caspase-9的表达量均显著或极显著下降(P<0.05或P<0.01)。说明鞣花酸的抗凋亡机制与抑制Bax、Bcl-2、caspase-3、caspase-8、caspase-9的表达有关。

     

    Abstract: To investigate the effects of ellagic acid on apoptosis of porcine intestinal epithelial cells(IPEC-J2) and the expression levels of related genes Bcl-2, Bax, caspase-3, caspase-8 and caspase-9, IPEC-J2 was treated with 20, 40, 60, 80 μg/mL ellagic acid for 24 hours, and 0 was added as control. The effects of different concentrations of ellagic acid on cell proliferation were detected by the method of tetramethylazazolate(MTT), the rate of cell apoptosis was determined by flow cytometry, the content of caspase-3 was determined by ELISA, and the mRNA expression of Bcl-2, Bax, caspase-8 and caspase-9 were determined by on intestinal epithelial cells real-time fluorescence quantitative PCR(qRT-PCR). Results show that the ellagic acid plays a protective role on intestinal epithelial cells, compared with 0 to add, different concentration of ellagic acid cells after 24 h, cell apoptosis rate, the expression levels of Bcl-2, Bax, caspase 3, caspase-8, and caspase-9 were significantly or extremely significantly decreased(P<0.05 or P<0.01). It indicated that the ellagic acid resistant mechanism of apoptosis and inhibition had related to the expression levels of Bax, Bcl-2, caspase 3, caspase-8, the expression of caspase-9.
