独龙鸡血细胞的形态观察
Morphological observation of blood cells of Dulong chicken

摘要: 为了观察和分析不同性别独龙鸡(Dulong chicken)血细胞的形态和大小差异,试验采集10只24周龄、体重为1.5~1.8 kg之间的健康独龙鸡(雌雄各半)血液,采用瑞氏-吉姆萨(Wright-Giemsa)染色法对血细胞进行染色,显微镜观察其形态并拍照保存图片,用Motic Images Plus 2.0 ML软件测量血细胞包括红细胞(未成熟红细胞和成熟红细胞)和白细胞(嗜中性粒细胞、嗜碱性粒细胞、嗜酸性粒细胞、单核细胞和大、小淋巴细胞)大小,人工计数法统计上述血细胞数量,并对上述指标进行性别差异独立t检验。结果表明:独龙鸡的成熟红细胞呈椭圆形,细胞核也呈椭圆形;未成熟红细胞呈卵圆形或椭圆形,细胞核呈椭圆形。嗜中性粒细胞呈圆形或椭圆形,细胞核多为分叶核,一般2~5叶;嗜酸性粒细胞呈圆形或椭圆形,细胞核分叶明显,通常为2叶;嗜碱性粒细胞呈圆形或椭圆形,细胞核分叶不明显。单核细胞体积较大,呈近圆形,细胞核不规则,多呈马蹄形、肾形等;大淋巴细胞整体略小于单核细胞,呈近圆形;小淋巴细胞整体呈近圆形,是白细胞中体积最小的细胞。疑似独龙鸡的凝血细胞常三五个聚集在一起,形成较大的块状结构,细胞核呈圆形、椭圆形或不规则形。雄性独龙鸡的成熟红细胞长径显著小于雌性(P<0.05),短径极显著小于雌性(P<0.01);但雄性独龙鸡的未成熟红细胞长径、短径均极显著大于雌性(P<0.01)。雄性独龙鸡嗜中性粒细胞和嗜碱性粒细胞的长径、短径均显著大于雌性(P<0.05);嗜酸性粒细胞和单核细胞的长径、短径性别差异均不显著(P>0.05);雄性独龙鸡大淋巴细胞和小淋巴细胞的长径、短径均极显著大于雌性(P<0.01)。独龙鸡的白细胞中小淋巴细胞数量最多,其次分别是大淋巴细胞、单核细胞、嗜中性粒细胞和嗜酸性粒细胞,嗜碱性粒细胞含量最少。独龙鸡的红细胞和白细胞数量性别差异均不显著(P>0.05)。说明独龙鸡红细胞的氧气运输能力在不同性别之间有差异,且雄性独龙鸡比雌性独龙鸡的免疫力更强。Abstract: In order to observe and analysis the differences in the morphology and size of blood cells of Dulong chicken of different sexes, blood samples were collected from 10 healthy Dulong chickens aged 24 weeks(half female and half male) with body weight of 1.5-1.8 kg. Wright-Giemsa staining method was used to stain blood cells, and the morphology was observed under a microscope and the pictures were taken for preservation. The size of blood cells(including red blood cells immature red blood cells and mature red blood cells and white blood cells basophils, eosinophils, neutrophils, monocytes and large and small lymphocytes) were measured with Motic Images Plus 2.0 ML software, and the number of the above blood cells was counted manually. The above indicators were tested by independent t test for sex differences. The results showed that the mature red blood cells of Dulong chicken and its nucleus were elliptic. Immature red blood cells were nearly ovoid or elliptic, and the nuclei were nearly elliptic. The neutrophils were round or elliptic, and the nuclei were mostly lobulated, generally 2 to 5 leaves. The eosinophils were round or elliptic, and the nuclei were obviously lobulated, usually 2 leaves. The basophils were round or elliptic, and the nuclei were not clearly lobulated. The monocytes were large in size and nearly round, and the nuclei were irregular, most of them were horseshoe-shaped and kidney-shaped. The large lymphocytes were slightly smaller than monocytes and were nearly round. The small lymphocytes were nearly round and were the smallest cell in white blood cells. The thrombocyte of Dulong chicken often gather together in three or five to form a larger block structure, and the nuclei were round, oval or irregular. The length diameter of mature red blood cells of male Dulong chicken were significantly lower than that of female(P<0.05), and the short diameter of mature red blood cells were extremely significantly smaller than that of female(P<0.01). However, the long and short diameters of immature red blood cells of male Dulong chicken were extremely significantly larger than that of female(P<0.01). The length diameter and short diameter of neutrophils and basophils in male Dulong chicken were significantly larger than those in female(P<0.05). There were no significant differences in the length diameter and short diameter of eosinophils and monocytes between different genders of Dulong chicken(P>0.05). The length diameter and short diameter of large and small lymphocytes in male Dulong chicken were extremely significantly larger than those in female(P<0.01). In the white blood cells of Dulong chicken, the number of small lymphocytes was the largest, followed by large lymphocytes, monocytes, neutrophils and eosinophils, and the content of basophils was the least. There was no significant difference in the number of red blood cell and white blood cell between different genders Dulong chicken(P>0.05). These results indicated that the oxygen transport capacity of red blood cell of Dulong chicken was different between males and females, and the immunity of male Dulong chicken was stronger than those of female Dulong chicken.
