Abstract:
In order to study fat mass and obesity-related gene(FTO) SNP polymorphism and their correlation with some slaughter traits in Tan sheep, 108 healthy adult Tan sheep were selected, and its anticoagulant were collected and genomic DNA was extracted. The polymorphism of FTO gene SNP locus(rs4784351 and rs7203051) were detected by using the method of PCR and DNA sequencing, and the correlation between the SNP loci and the slaughter indicators(living weight, carcass weight, kidney fat weight, gastrointestinal fat weight, tail fat weight) of Tan sheep was analyzed. The results showed that three genotypes(TT, TC and CC) were detected in 108 samples at rs4784351 locus, which were controlled by two alleles(T and C),and T was the dominant allele.The rs4784351 locus was significantly and extremely significantly correlated with carcass weight and tail fat weight of Tan sheep(P<0.05,P<0.01). The rs7203051 locus polymorphism was only detected in 19 Tan sheep, so no statistical analysis was performed. These results indicated that rs4784351 locus of FTO gene was significantly correlated with tail fat weight of Tan sheep, which could be used as an important reference molecular marker site for breeding selection of Tan sheep.
