基于流式细胞术的猪精液冷冻前质量控制研究

摘要: 为了探讨猪精液质量检测中相关指标的有效性,保障猪精子功能,试验首先利用精子自动分析系统对5头成年荣昌公猪新鲜精液、冷冻前精液(17℃保存2 d)、解冻精液进行检测;然后利用冻存精液开展人工授精试验,对精子功能进行验证;最后采用流式细胞术,以正常公猪冷冻前精液为对照,分析受精效果较差公猪新鲜精液的精子质膜完整性、线粒体活性及顶体完整性。结果表明:5头成年荣昌公猪新鲜精液的精子外形正常、密度与活力良好,符合相关标准要求;2~5号公猪能正常繁育,而使用1号公猪冻存精液配种的母猪均未怀孕;1号公猪活精子的质膜完整性(占比84.36%)与线粒体活性(占比40.00%)均高于对照组(72.43%和29.09%),但其活精子顶体完整性仅为41.74%,远远低于对照组的91.83%,揭示1号公猪是因精子顶体不完整导致的不育。说明在猪精液冷冻前仅检测精子外形、密度、活力并不能确保精子功能,还应结合流式细胞术对精子质膜完整性、线粒体活性及顶体完整性进行质量控制,以弥补传统指标检测的不足,才能保障养猪业健康可持续发展。Abstract: In order to investigate the validity of relevant indicators in semen quality detection and guarantee the function of pig sperm, the fresh semen, pre-freezing semen(stored at 17 ℃ for two days) and thawed semen of five adult Rongchang boars were detected by automatic sperm analysis system. Artificial insemination tests were then carried out using frozen semen and sperm function was verified. Finally, plasma membrane integrity, mitochondrial normal rate and acrosomal integrity of fresh semen of the boar with poor insemination were analyzed by flow cytometry with normal semen before freezing as control. The results showed that the sperm of five adult Rongchang boars were normal in shape, good in density and motility, which met the requirements of relevant standards. No. 2 to No. 5 boars could breed normally, but sows bred with frozen semen of No. 1 boar were not pregnant. The plasma membrane integrity(account for 84.36%) and mitochondrial activity(account for 40.00%) of live sperm of No. 1 boar were higher than those of the control group(72.43% and 29.09%), but the acrosomal integrity of live sperm of No. 1 boar was only 41.74%, which was much lower than that of the control group(91.83%), indicating that the incomplete acrosomal sperm of No. 1 boar led to infertility. The results indicated that only detecting the shape, density and vitality of sperm before freezing could not guarantee the function of sperm. The integrity of plasma membrane, normal rate of mitochondria and integrity of acrosome of sperm should be controlled by flow cytometry to make up for the deficiency of traditional index detection, so as to ensure the healthy and sustainable development of pig industry.
