DF-1细胞替代CEF细胞测定IBDV液抗原含量和血清中和抗体效价的研究
Study on DF-1 cells replacing CEF cells in the determination of antigen contents in IBDV fluid and serum neutralizing antibody titer

摘要: 为了探讨DF-1细胞替代鸡胚成纤维(chicken embryo fibroblast, CEF)细胞作为培养基质用于测定鸡传染性法氏囊病毒(Infectious bursal disease virus, IBDV)液抗原含量及血清中和抗体效价的可行性,试验以DF-1细胞和CEF细胞作为培养基质测定了6批次IBDV BJQ902株病毒液抗原含量和用不同剂量ND、IB、IBD三联灭活疫苗(La Sota株+M41株+BJQ902株)免疫SPF鸡血清和未免疫SPF鸡血清的中和抗体效价。结果表明:6批次病毒液病毒含量为1×107.7~1×108.1 TCID50/0.1 mL,两种细胞对同一批次病毒样品的测定结果一致,重复测定无差异;IBDV阳性血清样品中和抗体效价为8~17 lb,两种细胞对同一血清样品中和抗体效价的检测结果无差异。说明DF-1细胞可以替代CEF细胞作为培养基质,用于IBDV液抗原含量及血清中和抗体效价的测定。Abstract: In order to explore the feasibility of DF-1 cells replacing chicken embryo fibroblast(CEF) cells as a culture substrate for the determination of the antigen contents of Infectious bursal disease virus(IBDV) liquid and serum neutralizing antibody titer, in the experiment, DF-1 cells and CEF cells were used as culture substrates to determine the antigen contents of six batches of IBDV BJQ902 strain virus liquid, and neutralizing antibody titers of SPF chickens immunized with different doses of ND-IB-IBD(La Sota strain+M41 strain +BJQ902 strain) triple inactivated vaccine and serum of non immunized SPF chickens. The results showed that the antigen contents of the six batches of virus liquid were 1×107.7-1×108.1 TCID50/0.1 mL,and the results of the two kinds of cells on the same virus sample were consistent; and there was no difference between repeated determinations. The neutralizing antibody titers of IBDV-positive serum samples were 8-17 lb; there was no difference in the neutralizing antibody detection results of the same serum sample between the two kinds of cells. The results suggested that DF-1 cells could replace CEF cells for antigen contents determination of IBDV fluid and be used as the culture substrate for determination of serum neutralizing antibody titer.
