病毒活疫苗中沙门氏菌检测方法的改进

摘要: 为了改进病毒活疫苗中沙门氏菌检测方法,试验探究了增菌对检出率的影响,对沙门氏菌鉴别培养基进行了对比筛选,对改进的血清学检测方法进行了验证,并建立了分子生物学检测方法。结果表明:增菌可提高检出率,SS琼脂及HE琼脂较为适宜在检测中使用,血清学方法中用A~F群沙门氏菌属特异性诊断血清替代O多价1沙门氏菌因子血清是可行的,建立的分子生物学方法可用于最终鉴定病毒活疫苗中污染的杂菌。说明改进后的检测方法可提高沙门氏菌检出率,降低假阳性率,并能对污染的其他杂菌也进行种属鉴定。Abstract: In order to improve the detection method of Salmonella in live virus vaccine, this study explored the effect of enrichment on the detection rate, compared and screened Salmonella identification medium, verified the improved serological method, and established a molecular biology method. The results showed that the detection rate could be improved by bacteria enrichment. SS agar and HE agar were suitable for detection. In serological methods, it was feasible to replace O-multivalent-one Salmonella factor serum with A-F groups Salmonella specific diagnostic serum. The established molecular biological method could be used to finally identify the contaminated bacteria in live virus vaccine. The results suggested that the improved detection method could increase the detection rate of Salmonella, reduce the false positive rate, and could also identify the species of other contaminated bacteria
