Abstract:
The aim of this study was to reveal the histological characteristics of muscle fiber in different parts of breast muscle of Ningdu yellow chicken. Six 300-day-old Ningdu yellow hens were selected as the study subjects. The breast muscle on the right side of the outer layer(top, middle, bottom), the right side of the inner middle and the left side of the outer bottom were selected and recorded as group A, group B, group C, group D and group E, respectively. The conventional paraffin section method was used to determine the diameter and density of muscle fiber, number of fibers in muscle bundle, endomysium thickness and perimysium thickness. Meanwhile, the shear force values of the upper outer layer and the upper inner layer of the left pectoralis were also measured, and the correlation between the characteristic indexes of muscle fibers and the shear force values in all parts was analyzed. The results showed that there was no significant difference in the histological characteristics of muscle fibers among groups A, B, C,D and E(P>0.05), the number of fibers in muscle bundle in group D was significantly higher than that in the other four groups(P<0.05), the thickness of endomysium in group D was extremely significantly higher than that in group A, group B and group C(P<0.01), and significantly higher than group E(P<0.05). The shear force of outer layer of pectoralis was extremely significantly higher than inner(P<0.01). The muscle fiber diameter and perimysium thickness were extremely significantly negatively correlated with muscle fiber density(P<0.01). The diameter of muscle fiber and the thickness of endomysium were significantly positively correlated with the thickness of perimysium(P<0.05), and the number of fibers in the muscle bundle was significantly negatively correlated with the shear force(P<0.05). The results showed that there was no significant difference in the characteristics of muscle fiber in the outer part of the breast muscle of the same individual chicken, but there were differences in the characteristics of muscle fiber in the outer part of the breast muscle and the inner part of the breast muscle, suggesting that the sampling location in the breast muscle should be at the same area.
