一株青贮窖源丁酸梭菌的分离鉴定及其对小鼠的毒性和生长性能的影响

Isolation and identification of a strain of Clostridium butyricum derived from silage silos and its effect on toxicity and growth performance of mice

  • 摘要: 为了对紫花苜蓿青贮窖中的丁酸梭菌进行分离鉴定,并研究其对小鼠毒性和生长性能的影响,试验从青贮紫花苜蓿中分离培养丁酸梭菌,疑似菌株经形态学、生理生化以及16S rDNA序列鉴定后,分析阳性丁酸梭菌的最适生长温度和pH值,然后进行小鼠急性经口毒性试验将20只体重为18~22 g的SPF雄性昆明(KM)小鼠随机分为对照组和丁酸梭菌组,每组10只,丁酸梭菌组按2×1010 cfu/kg灌胃丁酸梭菌分离株稀释液,对照组灌胃等体积的生理盐水,每天9:00和15:00进行灌胃,预试期5 d,试验期10 d,试验前称重,记为初重,试验结束后颈椎脱臼法处死小鼠,称重,记为末重,取心脏、肝脏、脾脏、肾脏称重并计算各脏器系数)和其对小鼠生长性能影响试验(将120只体重为18~22 g的SPF雄性KM小鼠随机分为对照组、低剂量组、中剂量组、高剂量组、商品菌组,每组6个重复,每个重复4只,对照组灌胃生理盐水,低剂量组、中剂量组、高剂量组分别按2×10~6,2×10~8,2×1010 cfu/kg灌胃丁酸梭菌分离株稀释液,商品菌组按2×10~8 cfu/kg灌胃商品丁酸梭菌,每天9:00和15:00灌胃,预试期5 d,试验期28 d,于试验前和试验第14,28天称重,记录每天的耗料量、腹泻数和死亡数,计算0~14 d和15~28 d的平均日增重、平均日采食量、料重比、腹泻率和死亡率)。结果表明:从青贮紫花苜蓿中成功分离一株丁酸梭菌,命名为C3菌株,其最适生长温度在35~40℃之间、pH值在7.0~8.0之间。丁酸梭菌组与对照组小鼠均未发生中毒、死亡现象,且两组小鼠的初重、末重和心脏、肝脏、脾脏、肾脏的重量以及各脏器系数均差异不显著(P>0.05)。C3菌株灌胃14 d前,各组KM小鼠之间平均日增重、平均日采食量和料重比均差异不显著(P>0.05),腹泻率和死亡率均为0;灌胃14 d后,与对照组比,低、中、高剂量组和商品菌组可显著提高KM小鼠平均日增重(P<0.05),降低平均日采食量(P<0.05或P>0.05),以及显著降低料重比(P<0.05)。说明从青贮紫花苜蓿中成功分离到一株丁酸梭菌,其具有开发成为绿色、无毒饲料添加剂的潜在可能。

     

    Abstract: In order to study the isolation and identification of Clostridium butyricum in alfalfa silage silo and study its effect on toxicity and growth performance of mice. In this study, Clostridium butyricum was isolated and cultured from silage alfalfa. The suspected strains were identified by morphological, physiological and biochemical and 16 S rDNA sequence analysis. The optimal growth temperature and pH value of positive Clostridium butyricum were analyzed, and then the toxicity and growth performance of mice were tested. Acute oral toxicity test: 20 SPF male Kunming(KM) mice with body weight of 18 to 22 g were randomly divided into control group and Clostridium butyricum group, with 10 mice in each group. The Clostridium butyricum group was gavaged with 2×1010 cfu/kg Clostridium butyricum isolate diluent and the control group was given equal volume of normal saline by gavage at 9:00 and 15:00 every day. The pre-test lasted for 5 days and the experiment lasted for 10 days. Weighing before the test, denoted as initial weight. After the end of the experiment, the mice were killed by cervical dislocation, weighed and recorded as the final weight. The heart, liver, spleen and kidney were weighed and the coefficients of each organ were calculated. Growth performance test: 120 SPF male KM mice with body weight of 18 to 22 g were randomly divided into control group, low-dose group, medium-dose group, high-dose group and commercial bacteria group with 6 replicates in each group and 4 mice in each replicate. The control group was intragastric administered with normal saline, the low-dose group, medium-dose group and high-dose group were intragastric administered with 2×10~6,2×10~8,2×1010 cfu/kg Clostridium butyricum isolated diluent, respectively, and the commercial bacteria group was intragastric administered with 2×10~8 cfu/kg Clostridium butyricum at 9:00 and 15:00 every day. The pre-test lasted for 5 days, and the experiment lasted for 28 days. Weighing before the experiment and on the 14 th and 28 th day of the experiment, daily food consumption, diarrhea number and death number were recorded. The average daily gain, average daily feed intake, feed/gain, diarrhea rate and mortality were calculated on 0 to 14 days and 15 to 28 days. The results showed that a strain of Clostridium butyricum, named as strain C3, was successfully isolated and purified from silage alfalfa. The optimum growth temperature of Clostridium butyricum was between 35 ℃ and 40 ℃ and the pH value was about 7.0-8.0. No poisoning or death occurred between the Clostridium butyricum group and the control group, and there were no significant differences in the initial and final body weight, the weight of the heart, liver, spleen, kidney and the coefficient of each organ between the two groups(P>0.05). Zero to fourteen days after intragastric administration, there were no significant differences in average daily gain, average daily feed intake and feed/gain among KM mice(P>0.05), and the diarrhea rate and death rate were 0; 14-28 days of intragastric administration, compared to the control group, average daily gain in low, medium and high dose groups were significanty increased(P<0.05), average daily feed intake were reduced(P<0.05 or P>0.05),and feed/gain were significantly reduced(P<0.05). The results indicated that Clostridium butyricum C3 strain isolated from silage alfalfa had the potential to be developed as a green and non-toxic feed additive.
