鸭圆环病毒Ⅰ型与Ⅱ型Cap蛋白原核表达及免疫原性分析

Prokaryotic expression and immunogenicity analysis of Cap protein of Duck circovirus Ⅰ and Ⅱ

  • 摘要: 为了比较鸭圆环病毒(Duck circovirus, DuCV)Ⅰ型与Ⅱ型Cap蛋白免疫原性,试验分别选取DuCV-BJ株(HM162350.1,DuCVⅠ型)和DuCV-FJ株(EF370476.1,DuCVⅡ型)Cap基因作为参考序列,进行抗原表位预测,密码子优化与合成,克隆至pET-28a载体,将重组质粒转化至BL21(DE3)宿主菌中进行诱导表达,获得重组蛋白。重组蛋白纯化后免疫Balb/c小鼠,检测免疫小鼠体内特异性抗体IgG水平和细胞因子干扰素-γ(IFN-γ)、白细胞介素2(IL-2)、IL-4。结果表明:DuCV-BJ株和DuCV-FJ株Cap蛋白平均抗原指数分别为1.727和0.505,抗原表位区域存在差异。密码子优化后,密码子适应指数(CAI)由原来的0.89增长至1.00。重组质粒pET28a-BJ和pET28a-FJ均在大肠杆菌中成功表达;重组蛋白rCap-BJ和rCap-FJ免疫Balb/c小鼠后,均可诱导小鼠产生特异性抗体IgG,三免后rCap-FJ组、rCap-BJ组IgG水平极显著高于对照组(P<0.01),但rCap-FJ组与rCap-BJ组差异不显著(P>0.05);rCap-FJ组、rCap-BJ组IFN-γ、IL-2、IL-4水平极显著高于对照组(P<0.01)。说明重组蛋白rCap-BJ和rCap-FJ均能够刺激Balb/c小鼠机体产生特异性免疫应答,具有良好的免疫原性。

     

    Abstract: In order to compare the immunogenicity of Duck circovirus(DuCV) Ⅰ and Ⅱ Cap protain, the Cap gene of DuCV-BJ strain(HM162350.1, DuCVⅠ) and DuCV-FJ strain(EF370476.1, DuCV Ⅱ) were selected as reference sequences for antigen epitope prediction, codon optimization and synthesis, and cloned into pET-28 a vector which then was transformed into BL21(DE3) host bacteria for induced expression, and the recombinant proteins were obtained. Balb/c mice were immunized with the purified recombinant protein, and the level of specific antibody IgG and cytokines interferon-γ(IFN-γ), interleukin(IL)-2, and IL-4.The results showed that the average antigen index of Cap protein of DuCV-BJ strain and DuCV-FJ strain were 1.727 and 0.505, respectively. After codon optimization, the codon adaptation index(CIA) increased from 0.89 to 1.00. The recombinant plasmids pET28 a-BJ and pET28 a-FJ were successfully expressed in E. coli;Balb/c mice immunized with recombinant proteins rCap-BJ and rCap-FJ, both of which could induce mice to produce specific antibody IgG. After three immunization, IgG levels in rCap-FJ and rCap-BJ groups were very significantly higher than those in the control group(P<0.01), but there was no significant difference between rCap-FJ group and rCap-BJ group(P>0.05); the levels of IFN-γ, IL-2 and IL-4 in rCap-FJ and rCap-BJ groups were very significantly higher than those in the control group(P<0.01). The results suggested that the recombinant proteins rCap-BJ and rCap-FJ could stimulate Balb/c mice to produce specific immune response, and had good immunogenicity.
