Abstract:
In March 2021, Rabbit hemorrhagic disease virus(RHDV) infection was suspected in meat rabbits in a rabbit farm in Henan Province. In order to isolate and identify the infected strain from meat rabbits, and to study the characteristics of the isolated strain, the test dissected sick and dead rabbits being sent from the rabbit farm, and carried out bacteria and virus separation, RT-PCR detection, purity detection, hemagglutination test determination; and LD
50, traditional vaccine immunization rabbit challenge protection test and immunogenicity determination were further conducted. The results showed that the main manifestations of dead rabbits were extensive bleeding in liver, heart, lung and other parenchymal organs; no bacteria was isolated in diseased samples; RT-PCR detection identified the infected virus as RHDV. After injection of non-immunization healthy rabbits, the rabbits died within 48 hours, and the symptoms and necropsy lesions were similar to rabbit hemorrhagic disease. The isolated RHDV was named HN strain, which had no hemagglutination to human “O” red blood cells, and its LD
50 to rabbits was 1×10
-6.77/mL. Immunized rabbits after inactivation could resist the attack of RHDV HN strain. The results suggested that the meat rabbits in the rabbit farm were infected with RHDV, and a variant RHDV was successfully isolated and identified, which had good immunogenicity
