Abstract:
In order to establish a quick efficient method for the enrichment of Echinococcus eggs, this experiment discussed the setting of the key test conditions of flotation and centrifugation methods for the enrichment of Echinococcus eggs. In the flotation method, the enrichment rates of eggs were compared by using saturated solutions of sucrose, CaCl
2, NaCl, MgSO
4, ZnCl
2, and ZnSO
4 as floating solutions; and the optimal flotation durations(10,20,30,40,50,60,120,180 min)were screened out. In the centrifugation method, the enrichment rates of eggs were compared by using different centrifugation speeds(1 000-1 2000 r/min)and centrifugation durations(5,10,20,30 min), and the optimal combination of centrifugation speed and duration was explored. The results showed that when flotation method was used for the enrichment of eggs, the saturated CaCl
2 solution for 30 minutes was screened out, which could obtain the highest rate of the enrichment of eggs(42.1%). When centrifugation method was used for the enrichment of eggs, the parameter combination of 10 000 r/min and 10 min was used, and the optimal rate of the enrichment of eggs was obtained(62.2%). The results suggested that by optimizing the test condition parameters of flotation and centrifugation methods, the enrichment method of Echinococcus eggs could be optimized for the improvement of the enrichment of eggs.
