Abstract:
In order to improve the sperm motility before and after freezing of bovine straw frozen semen and improve the fertilization rate of frozen semen, semen quality indicators(semen volume, sperm motility, sperm density, number of frozen sperm produced, sperm recovery rate) before and after freezing were measured by stimulating the bulls with vaginal mucus from estrus cows, adding 1% Chinese herbal medicine additives in the diet, and thawing frozen semen at different temperatures and times, and the conditions that can improve the quality of frozen semen of Simmental bulls were screened. The results showed that the sperm motility of fresh sperm and frozen sperm, and the number of frozen semen available for production in the stud bulls smeared with vaginal mucus from estrus cows were significantly improved compared with stud bulls unsmeared with vaginal mucus from estrus cows(P<0.05),and the sperm density and semen volume of fresh sperm of each cattle were increased, but the differences were not significant(P>0.05). Adding Chinese herbal supplement to the diets of breeding bulls significantly improved the fresh sperm motility, frozen sperm motility and sperm density of the bulls(P<0.05), and the sperm recovery rate was also significantly improved, but the difference was not significant(P>0.05). The sperm motility was higher when thawed at 90 ℃ for 3-4 s, at 80 ℃ for 4-5 s, at 70 ℃ for 4-6 s, at 60 ℃ for 8-9 s, and at 50 ℃ for 20-30 s. The sperm motility was the highest and stable when the bovine frozen semen was thawed at 70 ℃ for 4-6 s and 80 ℃ for 4-5 s. The results indicated that the application of vaginal mucus stimulation of estrus cows, feeding of Chinese herbal medicine additives and reasonable thawing temperature and time of frozen bovine semen could improve the sperm motility of frozen bovine semen.
