Abstract:
In order to understand the relationship between the proliferation of Senecavirus A(SVA) and IL-6, in the experiment, PCR identification, indirect immunofluorescence, whole genome sequence evolutionary analysis and other methods were used to determine the infection and genetic evolution of suspected samples, and fluorescence quantitative assay and siRNA interference assay were used to determine the effect of IL-6 on SVA proliferation.The results showed that the SVA fragment of expected size was amplified, and the PK15 cell showed obvious cytopathic effect and specific green fluorescence which inoculated with vesicle filtrate. The isolated strain was named CHgx. The strain was in the same branch with the domestic USA/GBI29/2015-like strain. SVA CHgx strain could significantly promote the expression of IL-6 in SVA-infected PK15 cells(P<0.001), and the interference of IL-6 expression could significantly inhibit SVA replication in cells(P<0.001). These results suggesed the inhibition of interference of IL-6 expression against the proliferation of SVA.
