干扰IL-6基因表达抑制A型塞内卡病毒的增殖

Inhibition of interference of IL-6 gene expression against the proliferation of Senecavirus A

  • 摘要: 为了了解A型塞内卡病毒(SVA)的增殖和IL-6的关系,试验采用PCR鉴定、间接免疫荧光、全基因组序列进化分析等方法确定疑似病料感染和遗传演化情况,同时采用荧光定量和siRNA干扰试验确定IL-6对SVA增殖的影响。结果表明:PCR扩增得到预期大小的SVA片段,接种囊泡过滤液的PK15细胞出现明显的细胞病变效应,且出现特异的绿色荧光,将分离毒株命名为CHgx。该毒株与国内USA/GBI29/2015-like毒株处于同一分支。SVA CHgx毒株在感染PK15细胞后能极显著促进IL-6的表达(P<0.001),干扰细胞中IL-6表达后极显著抑制SVA的复制(P<0.001)。说明干扰IL-6的表达可抑制SVA增殖。

     

    Abstract: In order to understand the relationship between the proliferation of Senecavirus A(SVA) and IL-6, in the experiment, PCR identification, indirect immunofluorescence, whole genome sequence evolutionary analysis and other methods were used to determine the infection and genetic evolution of suspected samples, and fluorescence quantitative assay and siRNA interference assay were used to determine the effect of IL-6 on SVA proliferation.The results showed that the SVA fragment of expected size was amplified, and the PK15 cell showed obvious cytopathic effect and specific green fluorescence which inoculated with vesicle filtrate. The isolated strain was named CHgx. The strain was in the same branch with the domestic USA/GBI29/2015-like strain. SVA CHgx strain could significantly promote the expression of IL-6 in SVA-infected PK15 cells(P<0.001), and the interference of IL-6 expression could significantly inhibit SVA replication in cells(P<0.001). These results suggesed the inhibition of interference of IL-6 expression against the proliferation of SVA.
