Abstract:
In order to establish a high-performance liquid chromatographic(HPLC) fingerprint combined with quantitative analysis of multi-components by single marker(QAMS) to evaluate the quality of Dai medicine prescription Huang’s Lanwenkang(DMPHL), in this experiment, HPLC was used to analyze the components of 11 batches of DMPHL compounds; the obtained chromatographic information was imported into the “Chinese medicine chromatography fingerprint similarity evaluation system” 2012 edition for analysis; the common peak identification was identified by the reference substances, and the HPLC fingerprint of DMPHL was established. Using chrysophanol as an internal reference, QAMS method was established which could simultaneously determine the contents of chlorogenic acid, palmatine hydrochloride, berberine hydrochloride, chrysophanic acid, ethyl p-methoxycinnamate, emodin and chrysophanol. At the same time, the external standard method(ESM) was used to verify the accuracy of the determination results of the QAMS method. The results showed that the HPLC fingerprints of 11 batches of DMPHL had a total of 24 peaks, and the similarity of DMPHL fingerprints of different batches was above 0.900. Chrysophanol was used as an internal reference, the relative correction factors of chlorogenic acid, palmatine hydrochloride, berberine hydrochloride, chrysophanic acid, ethyl p-methoxycinnamate, emodin were calculated to be 2.017 0, 0.788 5, 0.872 5, 7.926 4, 2.713 1 and 0.699 2, respectively. The RSD value of the determination results of the QAMS method and the external standard method was less than 3.00%, and there was no obvious difference between the determination results of the two methods, which suggested that the HPLC fingerprint established in the experiment combined with QAMS method was suitable for the quality evaluation of DMPHL.
