Abstract:
In order to explore the effects of TYRP1 gene on the coat color of Chinese domestic donkeys, in the experiment, blood samples were collected from 410 Chinese donkeys with different coat colors, and polymerase chain reaction and single-stranded conformation polymorphism(PCR-SSCP) methods were used to detect single nucleotide polymorphism(SNP) sites in the second intron of the TYRP1 gene of Chinese donkeys. Individuals of Chinese donkeys with different coat colors were genotyped and genotype frequencies and allele frequencies were calculated; the relationship between polymorphisms in the second intron of the TYRP1 gene and different coat colors of Chinese national donkeys was analyzed. The results showed that one SNP locus(g.1 702A→G) was detected in the second intron of the TYRP1 gene of the Chinese domestic donkeys; this locus contained three genotypes, AA, AG and GG. The dominant allele was A in black, black three-pink, green three-pink, gray three-pink, white and albino domestic donkeys; black, black three-pink and green three-pink all had the highest frequency of the AA genotype and the lowest frequency of the GG genotype; gray three-pink had the highest frequency of the AA genotype and the lowest frequency of the AG genotype(0). Among the black, black three-pink, green three-pink and grey three-pink donkeys, the AA genotype frequency was the highest for gray three-pink, followed by green three-pink, black three-pink, and black in that order; the frequency of GG genotypes of domestic donkeys with predominantly black coat color(black, black three-pink, and green three-pink) was lower than that of donkeys with predominantly grey coat color(grey three-pink); black three-pink and green three-pink with a mixture of white hairs had a lower frequency of GG genotypes than that of purely black. There were two white domestic donkeys and two albino domestic donkeys each in the sample, all of which were genotyped as AA pure. The results indicated that the AA genotype frequency of g.1 702A →G of TYRP1 gene decreased with the increase of the proportion of black coat in Chinese domestic donkeys, suggesting that the AA genotype frequency of G1,702A→G in TyrP1 gene might be related to the hair color traits of Chinese domestic donkeys.
