Abstract:
To investigate the expression difference of pyruvate dehydrogenase kinase 4(PDK4) gene in different tissues of sheep and the effects of gene exons polymorphism on meat quality traits, 45 healthy 8-month-old Shuangqian rams were selected as the research subjects in the experiment. After slaughter, different tissues(heart, liver, spleen, lung, kidney, intestine, longissimus muscle) were collected. The single nucleotide polymorphism(SNP) in the exon of PDK4 gene was detected by a combination method of PCR and Sanger sequencing, and then associated with meat quality data. The results showed that the PDK4 gene was widely distributed in various tissues, and its relative expression was the highest in duodenum tissues. There were two SNP in the first exon of the PDK4 gene(T>C mutation at Chr 4. 14723148 and T>C mutation at Chr 4. 14723208). There were two genotypes at Chr 4. 14723148 position, which were CC and CT. There were three genotypes at Chr 4. 14723208 position, namely TT, TC, and CC. The distribution of mutated genotypes followed Hardy Weinberg’s law, indicating that the mutated individual was in a stable state in the population. The polymorphism information content was 0.94 and 0.61, respectively, both of which were highly polymorphic(P>0.5). The CC genotype at Chr 4. 14723148 locus was the dominant genotype, and its individual drip loss rate was significantly lower than that of the CT genotype(P<0.05). The experimental results indicated that the Chr4. 14723148 SNP locus was stable in the sheep population, and the dominant gene CC type had highly water holding capacity that affected meat quality traits.
