Abstract:
In order to solve the problem of the diminished quality in the frozen sperm of Chongming white goats, semen samples from eight well-proportioned Chongming white goats aged 24 to 26 months were obtained and mixed to eliminate individual differences after qualified detection. Semen was programmed to freeze after treatment with basal diluent without angelica polysaccharide(ASP)or with 150,300,600,and 1 200 μg/mL ASP.Sperm quality parameters(viability, motility, plasma membrane integrity, acrosome integrity, mitochondrial activity),relevant oxidative indices(malondialdehyde(MDA),reactive oxygen species(ROS),superoxide dismutase(SOD),glutathione peroxidase(GPH-Px),nitric oxide(NO)content, total antioxidant capacity(T-AOC),catalase(CAT)),and sperm DNA methylation level were evaluated; the optimal concentration of ASP was determined, and the correlation between sperm DNA methylation 5-MC level and sperm quality parameters was analyzed.The results showed that viability, motility, plasma membrane integrity and mitochondrial activity of sperm in the 600 μg/mL ASP group were significantly higher than those in the group without ASP(P<0.05),and the acrosome integrity of sperm in the 300 μg/mL ASP group was significantly higher than that in the group without ASP(P<0.05).The SOD and CAT activities and T-AOC values of sperm in the 600 μg/mL ASP group were significant higher than those in the group without ASP(P<0.05),but ROS and MDA values of sperm in the 600 μg/mL ASP group were significantly lower than those in the group without ASP(P<0.05).The DNA methylation level of sperm in the 300 and 600 μg/mL ASP groups was significant lower than that in the group without ASP(P<0.05).Sperm DNA methylation 5-MC level was significantly negatively correlated with sperm motility, viability, plasma membrane integrity, acrosome integrity and mitochondrial activity(P<0.01), and R=-0.983,-0.892,-0.992,-0.732,-0.825. The results indicated that the addition of 600 μg/mL ASP in basal diluent was the most appropriate for the cryopreservation of Chongming white goat semen.
