4个羊草品种(系)耐盐性评价

Evaluation on salt tolerance of four Leymus chinensis varieties (lines)

  • 摘要: 为了探究不同羊草品种(系)耐盐性,试验以JM007、JM009、吉生1号和东北羊草为材料,分析不同浓度(200,400,600 mmol/L) NaCl胁迫对其萌发期萌发指标和幼苗期生理指标的影响,以纯化水/Hoagland营养液为对照(CK),并利用主成分分析法、相关性分析、隶属函数法等方法对各单项指标的耐盐系数进行分析,综合评价羊草品种(系)耐盐性。结果表明:羊草萌发期和幼苗期的各项指标均受到不同浓度NaCl胁迫的影响,但不同指标对NaCl胁迫的敏感度存在差异。随着NaCl浓度升高,羊草种子的相对发芽率、相对发芽势、相对发芽指数、相对种子活力、相对胚芽长和相对胚根长均呈逐渐下降趋势,此外NaCl胁迫对羊草胚芽长的抑制作用大于胚根长。与CK相比,不同浓度NaCl胁迫下4种羊草幼苗MDA、Pro含量均显著增加(P<0.05),JM007羊草幼苗SOD活性呈逐渐上升趋势,JM009、吉生1号和东北羊草幼苗SOD活性呈先上升后下降趋势。除东北羊草外,其他3种羊草幼苗POD、CAT活性相较CK均显著增加(P<0.05);与CK相比,JM007和JM009羊草幼苗SS含量均呈逐渐上升趋势,吉生1号羊草和东北羊草幼苗SS含量呈现先平稳后骤升最后下降的趋势。按综合评价(D)值排序,萌发期为JM009>JM007>吉生1号>东北羊草,幼苗期为JM007>吉生1号> JM009>东北羊草。说明JM009羊草种子在萌发期耐盐能力较强,JM007羊草则在幼苗期耐盐能力较强。

     

    Abstract: In order to explore the salt tolerance of different varieties(lines) of Leymus chinensis(L. chinensis), the effects of NaCl stress at different concentrations(200, 400, and 600 mmol/L) on germination and physiological indexes at seedling stage were analyzed in JM007, JM009, Jisheng 1, and Northeast L. chinensis. Using purified water/Hoagland solution as the control(CK), the salt tolerance coefficient of each single index was analyzed by principal component analysis, relationship analysis and membership function method, and the salt tolerance of L. chinensis was comprehensively evaluated. The results showed that all the indices in germination and seedling stages of L. chinensis were affected by NaCl stress, but the sensitivity of different indices to NaCl stress was different. With the increase of NaCl concentration, the relative germination rate, relative germination potential, relative germination index, relative vitality index, relative embryo length and relative radicle length of seeds decreased gradually. In addition, the inhibition effects of NaCl stress on the germination growth of seedlings was greater than that of radicle length. Compared with CK, the contents of MDA and Pro in four L. chinensis seedlings under different NaCl concentrations were significantly increased(P<0.05); the SOD activity of JM007 seedlings increased gradually, and the SOD activity of JM009, Jisheng No.1 and Northeast L. chinensis seedlings increased first and then decreased. Except for Northeast L. chinensis, the POD and CAT activities of the other three L. chinensis seedlings were significantly increased compared with CK(P<0.05). Compared with CK, the SS content of JM007 and JM009 seedlings increased gradually, and the SS content of Jisheng 1 and Northeast L. chinensis seedlings showed a steady development first, then a sharp rise and finally a decline. According to the comprehensive membership function value evaluation(D) value, the order of salt tolerance strength was: JM009>JM007> Jisheng No.1 > Northeast L. chinensis at germination stage, and JM007> Jisheng No.1 > JM009> Northeast L. chinensis at seedling stage. The results indicated that JM009 had a strong salt tolerance at the germination stage, while JM007 had a strong salt tolerance at the seedling stage.
