Abstract:
To study the structure and function of the chicken FOXL2 gene, in this study, the FOXL2 gene coding region sequence of Guangxi indigenous chicken was cloned. The mutation site, homology, and evolutionary distance with other species, as well as the expression level of FOXL2 gene in ovarian tissues of hens in high-yield and low-yield groups were analyzed. The results showed that there was a FOXL2 gene coding region sequence of 918 bp in Guangxi indigenous chicken, encoding 305 amino acids, with one missense mutation and two synonymous mutations.The homology analysis showed that the homology of FOXL2 gene of Guangxi indigenous chicken with FOXL2 gene of Gallus gallus, Phasianus colchicus, Anas platyrhynchos, Columba livia, Homo sapiens, and Mus musculus were 99.7%, 98.8%, 95.1%, 92.0%, 75.7%, and 75.5%. Phylogenetic tree analysis showed that FOXL2 gene in Guangxi indigenous chicken had the closest relationship with FOXL2 gene in Gallus gallus and the farthest relationship with FOXL2 gene in Mus musculus. The expression level of FOXL2 gene in ovaries tissues of high-yield and low-yield hens showed that the expression level of FOXL2 gene in the high-yield group was significantly higher than that in the low-yield group(P<0.05). The results of this study suggested that the sequence of FOXL2 gene in chickens was relatively conservative and of functions for maintaining ovarian function and improving egg production.
