基于转录组筛选黑安格斯牛肉质性状关键调控基因

Screening of Key Regulatory Genes for Beef Quality Traits in Black Angus Based on Transcriptome Sequencing

  • 摘要: 为了对黑安格斯牛肉质性状关键调控基因进行筛选,本研究分别以3头24月龄的黑安格斯牛和西门塔尔牛背最长肌为试验材料,在运用Illumina HiSeqTM技术对所有样品进行转录组测序后利用生物信息学方法和相关软件对测序结果进行归类分析。通过分析得到黑安格斯牛与西门塔尔样本间的差异表达基因有409个,其中上调基因115个,下调基因294个。进一步分析发现与肉质性状相关的基因PPARD表达量在黑安格斯中显著高于西门塔尔(P<0.01),对此运用RT-qPCR技术进行验证得到的实验结果与转录组结果一致。这些研究结果不仅表明黑安格斯牛中的PPARD表达量比西门塔尔牛的高,还说明该基因对牛的肉质具有一定的调控作用,也对宁夏地区安格斯牛的选育及核心安格斯牛种群的建设具有一定的参考价值。

     

    Abstract: In order to screen the key regulatory genes of meat quality traits in black Angus cattle, the longissimus dorsi muscles of 3 24-month-old black Angus cattle and Simmental cattle were used as experimental materials. Illumina HiSeqTMtechnique was used to sequence all samples and bioinformatics methods and related software were used to classify and analyze the sequencing results. The differentially expressed genes between black Angus cattle and Simmental samples were 409, of which 115 were up-regulated and 294 were down-regulated. Further analysis showed that the expression of PPARD, which was related to meat quality traits, was significantly higher in Black Angus than that of Simmental(P<0.01). The experimental results were consistent with those of the transcriptional group by RT-qPCR. These results not only showed that the expression of PPARD in black Angus cattle was higher than that in Simmental cattle, but also indicated that the gene had a certain regulatory effect on beef quality. It also has certain reference value for the breeding of Angus cattle and the construction of core Angus cattle population in Ningxia.
