密斑刺鲀igf2基因的克隆及表达分析

Cloning and Expression Analysis of Igf2 Gene in Diodon hystrix

  • 摘要: 胰岛素样生长因子2 (insuline-like growth factors 2, IGF2)在控制脊椎动物的生长、发育、繁殖、代谢和渗透调节等方面发挥着关键作用。本研究利用分子克隆技术分离并鉴定了密斑刺鲀(Diodon hystrix) igf2基因cDNA序列738 bp,其中开放阅读框全长645 bp,可编码214个氨基酸残基。多重序列比对分析发现,密斑刺鲀IGF2氨基酸序列与其他鱼类的IGF2具有较高的同源性,显示出序列进化的保守性。组织分布结果表明,igf2基因在肝脏和脑组织中表达量较高,而在肾脏以及精巢中没有检测到表达信号,表明igf2基因具有显著的组织表达特异性。本研究结果为深入研究密斑刺鲀的igf2基因在生长发育中的调控机理提供研究基础。

     

    Abstract: Insulin-like growth factor 2(IGF2) plays a key role in the growth, development, reproduction, metabolism and osmotic regulation in vertebrates. In this study, the Diodon hystrix igf2 cDNA sequence of 738 bp was identified by molecular cloning technology, and contains the full-length ORF of 645 bp, encoding 214 amino acids.Multiple sequence alignment analysis revealed that the IGF2 in D. hystrix had high homology with other fishes,suggesting that IGF2 was conservative in teleost. The results of tissue distribution showed that the igf2 gene was mainly expressed in liver and brain, and was not expressed in kidney and testis. The results showed that the igf2 gene had significant tissue expression specificity. This study provided a basis for in-depth study of the regulatory mechanism of igf2 gene in growth and development.
