弓背青鳉zglp1基因的克隆和表达分析

Molecular Cloning and Expression Analysis of zglp1 from Oryzias curvinotus

  • 摘要: Zinc finger GATA like protein-1 (ZGLP1)是位于细胞核内的一种核转录因子结合蛋白,在哺乳动物性腺发育过程中调控生殖细胞和体细胞的相互作用。为了研究ZGLP1在鱼类中的功能,我们以弓背青鳉(Oryzias curvinotus)为研究对象,对弓背青鳉zglp1基因进行了PCR扩增、分子克隆、序列特征分析,并对其时空表达模式进行了反转录-PCR (reverse-transcription PCR, RT-PCR)检测。结果显示,在弓背青鳉卵巢组织cDNA中扩增获得了zglp1两种转录产物(orcuzglp1x1和orcuzglp1x2)。orcuzglp1x1长1 739 bp,包含一个1 665 bp的开放阅读框(ORF),预测编码蛋白含554个氨基酸(aa),蛋白分子量为61.40 kDa;orcuzglp1x2长1 629 bp,ORF长1 407 bp,编码蛋白含468 aa,蛋白分子量为51.81 kDa。orcuzglp1x2相对于orcuzglp1x1在N端缺失86 aa,orcuzglp1s在C端含有两个锌指结构(Znf1, Znf2),Znf1和典型的GATA锌指DNA结合基序高度同源。多序列比对结果显示Znf1在物种间较为保守,而Znf2的保守性较低。系统进化树分析结果表明Zglp1在进化上和物种进化保持一致。RT-PCR结果显示orcuzglp1s主要表达于成熟卵巢组织以及胚胎发育早期(卵裂期)。综上所述,弓背青鳉zglp1可能在卵子发生及胚胎卵裂过程中发挥一定作用。

     

    Abstract: Zinc finger GATA like protein-1(ZGLP1) is a nuclear zinc finger protein that regulates the interaction between germ cells and somatic cells during gonad developmental process in mammals. In order to study the function of ZGLP1 in fish, we took Oryzias curvinotus as the research object and carried out PCR amplification,molecular cloning and sequence characterization analysis of the zglp1 gene, and the spatiotemporal expression pattern was detected by reverse transcription-PCR. The results showed that two zglp1 transcripts(orcuzglp1 x1 and orcuzglp1 x2) were amplified from cDNA of O. curvinotus ovary tissue. Orcuzglp1 x1 was 1 739 bp in lengthand contained an open reading frame of 1 665 bp. It was predicted that the encoded protein contained 554 amino acids and the molecular weight of the protein was 61.40 kDa. Orcuzglp1 x2 was 1 629 bp in length, and the length of the open reading frame was 1 407 bp. The encoding protein contained 468 aa, and the molecular weight of the protein was51.81 kDa. Orcuzglp1 x2 had a deletion of 86 aa at the N terminal compared with Orcuzglp1 x1, and orcuzglp1 s had two zinc finger structures(Znf1, Znf2) at the C terminal. Znf1 was highly homology with the typical GATA zinc finger DNA binding motif. The results of multi-sequence alignment showed that Znf1 was more conserved among species,while Znf2 was less conserved. Phylogenetic tree analysis showed that Zglp1 was s consistent with species evolution.RT-PCR results showed that orcuzglp1 s was mainly expressed in mature ovarian tissues and early embryonic development(cleavage stage). In conclusion, the O. curvinotus zglp1 may play a role in oogenesis and embryo cleavage.
