白花蛇舌草提取物诱导人胃癌MKN-45细胞凋亡

摘要: 为探究白花蛇舌草提取物对人胃癌MKN-45细胞凋亡的影响。用不同浓度的白花蛇舌草提取物处理MKN-45细胞,然后在光学显微镜和激光共聚焦显微镜下观察数量和形态。流式细胞仪检测细胞凋亡率,RT-qPCR和Western Blot检测凋亡相关基因Bax和Bcl-2的m RNA和蛋白表达水平。结果表明,不同浓度的白花蛇舌草提取物处理48 h后,在显微镜下观察到细胞体积缩小、细胞核裂解和染色质形态变化。细胞凋亡率随着药物浓度的增加而增加,用30μg/mL白花蛇舌草提取物处理后细胞凋亡率达到23.6%,Bax基因表达水平显著增加,Bcl-2基因表达水平显著降低。综上所述,白花蛇舌草提取物可诱导人胃癌MKN-45细胞凋亡,具有潜在的医学和药用价值。Abstract: To invest igat e the effect of Hedyotis diffusa extract on apoptosis of human gastric cancer MKN-45 cells. The MKN-45 cells were treated with different concentrations of Hedyotis diffusa extract, and then the quantity and morphology were observed under light microscopy and laser confocal microscopy. The apoptosis rate was detected by flow cytometry. The mRNA and protein expression levels of apoptosis-related genes Bax and Bcl-2 were detected by RT-qPCR and Western Blot. The results showed that after treatment with different concentrations of Hedyotis diffusa extract for 48 hours, the cell volume shrinkage, nucleus cleavage and chromatin morphological changes were observed under the microscope. The apoptosis rate increased with the increase of drug concentration,and the apoptotic rate reached 23.6% after treated with 30 μg/mL of Hedyotis diffusa extract the expression level of Bax gene significantly increased, and Bcl-2 significantly decreased. In summary, Hedyotis diffusa extract can induce apoptosis of human gastric cancer MKN-45 cells, which has potential medical and medicinal value.
