TGF-β Signaling Pathway-mediated Related Diseases and Targeted Therapy

  • 摘要: TGF-β信号通路是一个由众多细胞因子组成的大家族,调节着包括细胞生长、上皮-间质转化、迁移、分化以及凋亡等在内的许多细胞功能,在组织与器官的正常生长、胚胎发育等过程中起着关键作用。一旦TGF-β信号传导过程发生异常,随之而来的就是一系列发育缺陷和疾病的产生,如肿瘤的发生,组织器官的纤维化,结缔组织以及骨骼疾病等。通过对TGF-β信号通路在不同疾病发生中的作用进行研究,目前已经开发了许多靶向此通路的治疗策略,如单克隆抗体、受体激酶抑制剂和反义寡聚核苷酸等。本综述总结了TGF-β信号通路在不同疾病发生中的作用,并对最新的靶向此信号通路的治疗策略进行讨论。

     

    Abstract: The TGF-β signaling pathway is a large family composed of numerous cytokines, which regulate many cellular functions, including cell growth, epithelial-mesenchymal transition, migration, differentiation and apoptosis, and plays a key role in the normal growth of tissues and organs, embryonic development and carcinogenesis.Once abnormal TGF-β signal transmission process occurs, a series of developmental defects and diseases are produced, such as occurrence of tumor, fibrosis of tissues and organs, connective tissue and bone diseases. By studying the role of TGF-β signaling pathway in the occurrence of different diseases, many therapeutic strategies targeting this pathway have been developed, such as monoclonal antibodies, receptor kinase inhibitors and antisense oligonucleotides. This review summarized the role of TGF-β signaling pathway in the occurrence of different diseases, and discussed the latest therapeutic strategies targeting this signaling pathway.
