小鳞拟松鲷3个养殖群体微卫星遗传多样性分析

Analysis of Microsatellite Genetic Diversity in Three Cultured Populations of Datnioides pulcher

  • 摘要: 通过聚丙烯酰胺凝胶电泳法筛选微卫星引物,从本实验室已筛选的20对微卫星多态性引物中挑选出7对特异性较强的多态性引物,对小鳞拟松鲷(Datnioides pulcher) 3个引进群体进行微卫星遗传多样性分析。结果显示所检测的7个微卫星位点共检测到31个等位基因,每个位点等位基因数3~7个,平均等位基因数(Na)为4.429,平均有效等位基因数(We)为3.122,平均观测杂合度(Ho)为0.668,平均期望杂合度(He)为0.655;3个引进群体的平均等位基因数(Na)分别为5.000,4.500和4.140,平均观测杂合度(Ho)分别为0.700,0.625和0.636,平均期望杂合度(He)分别为0.695,0.635和0.635。先达如湖群体(XDR)、坤甸群体(KD)、巨港群体(JG) 3个群体的PIC值由大到小依次为:0.630,0.574,0.540。可见3个群体的遗传多样性水平较高,各群体遗传距离较远,存在一定程度的遗传分化。先达如湖群体(XDR)的遗传多样性较高,作为选育群体具有一定的遗传潜力。

     

    Abstract: Microsatellite primers were screened by polyacrylamide gel electrophoresis, and 7 pairs of highly specific polymorphic primers were selected from the 20 pairs of microsatellite polymorphic primers screened in our laboratory, and microsatellite genetic diversity analysis was conducted on 3 populations of introduced groups of Datnioides pulcher. The results showed that a total of 31 alleles were detected in the 7 microsatellite loci detected,and the number of alleles(Na) at each loci ranged from 3 to 7. The average number of alleles was 4.429, the average number of effective alleles(Ne)was 3.122, the average observed heterozygosity(Ho) was 0.668, and the average expected heterozygosity(He) was 0.655. The mean number of alleles(Na) of the three introduced populations was 5.000, 4.500 and 4.140, the mean observed heterozygosity(Ho) was 0.700, 0.625 and 0.636, and the mean expected heterozygosity(He) was 0.695, 0.635 and 0.635, respectively. The PIC values of XDR, KD and JG were0.630, 0.574 and 0.540 respectively. It can be seen that the genetic diversity of the three populations is high, and the genetic distance of each population is far, so there is a certain degree of genetic differentiation. The genetic diversity of XDR population is high, so it has certain genetic potential as a selective breeding population.
