甘松饮改善细胞外基质堆积延缓糖尿病肾病的研究

Diabetic Kidney Disease Suppression by Gan-song Yin Improving Extracellular Matrix Accumulation

  • 摘要: 研究甘松饮(Gan-song Yin, HGY)对糖尿病肾病(diabetic kidneys disease, DKD)大鼠(Rattus norvegicus)肾脏FN、Col1α1和Col4α1基因及蛋白表达的影响。制备糖尿病肾病大鼠模型,分为正常对照组(Normal)、DKD模型组(DKD)、格列喹酮对照组(DKD+Gli, 10 mg/kg)、HGY高剂量组(DKD+HGY, 10 g/kg)及HGY低剂量组(DKD+HGY, 5 g/kg)。采用Western Blot法、免疫组化法检测DKD大鼠肾脏组织FN、ColⅠ、ColⅣ的表达蛋白水平,实时荧光定量PCR法检测DKD大鼠肾脏FN、Col1α1和Col4α1基因表达水平。与DKD比较,HGY可以显著降低DKD模型大鼠肾脏组织FN、ColⅠ、ColⅣ的表达蛋白水平以及FN、Col1α1、Col4α1的基因表达水平(P<0.05),且HGY与格列喹酮的调控作用基本接近。说明HGY可以下调早期DKD大鼠肾脏组织FN、ColⅠ、ColⅣ的表达蛋白水平以及FN、Col1α1、Col4α1的基因表达水平,可能是通过改善肾脏细胞外基质(extracellular matrix, ECM)堆积从而延缓DKD发展。

     

    Abstract: To study the effect of Gan-song Yin(HGY) on the expression of FN, Col1α1 and Col4α1 genes and proteins in the rats model of diabetic kidneys disease(DKD). Rat models of diabetic kidney disease were prepared and divided into normal control group(Normal), DKD model group(DKD), gliquidone control group(DKD +Gli,10 mg/kg), HGY high dose group(DKD+HGY, 10 g/kg) and HGY low-dose group(DKD+HGY, 5 g/kg). Western Blot and immunohistochemical methods were used to detect the protein expression levels of FN, ColⅠ, ColⅣ in DKD rat, and real-time fluorescence quantitative PCR method was used to detect the gene expression levels of FN,Col1α1 and Col4α1 genes in DKD rat. Compared with DKD, HGY can significantly reduce the protein levels of FN,ColⅠ, ColⅣ and the gene expression levels of FN, Col1α1, Col4α1 in DKD rat(P<0.05), and HGY is basically similar to gliquinone in regulation. HGY can down-regulate the protein levels of FN, ColⅠ, ColⅣ and the gene expression levels of FN, Col1α1, Col4α1 in early DKD rats, which may delay the development of DKD by improving extracellular matrix(ECM) accumulation.
