黄毛草莓FnERF017基因和启动子克隆及表达分析

Cloning and Expression Analysis of Gene and Promoter of FnERF017 in Fragaria nilgerrensis Schidl.

  • 摘要: ERF在植物的生长发育和生物胁迫响应中起着重要作用。为进一步研究草莓ERF基因的功能,以黄毛草莓叶片为材料,通过RT-PCR技术克隆获得1个FnERF017基因。基因序列及分子特征分析结果表明,FnERF017基因的cDNA全长序列为651 bp,编码216个氨基酸,具有一个AP2/ERF保守结构域,其编码产物为亲水性不稳定蛋白;聚类分析结果显示,黄毛草莓FnERF017蛋白与森林草莓的ERF蛋白具有高度同源性;胶孢炭疽菌(Colletotrichum gloeosporiodes)胁迫表达特征分析表明,Fn ERF017在胶孢炭疽菌胁迫条件下呈上调表达,表明其正向响应胶孢炭疽菌胁迫;上游启动子顺式作用元件预测显示,启动子pFnERF107序列中含有胁迫响应元件及激素响应元件等多种类型的顺式作用元件。研究结果为进一步研究FnERF017的基因功能提供基础。

     

    Abstract: ERF plays an important role in plant growth and development and in response to biotic stress. To further research the function of Fragaria nilgerrensis ERF genes, the F. nilgerrensis leaves were used to clone the gene FnERF017 by RT-PCR. The analysis of gene sequence and molecular characteristics showed that the full-length cDNA sequence of Fn ERF017 was 651 bp, encoding 216 amino acids with a conserved AP2/ERF domain. FnERF017 protein was a hydrophilic unstable protein. The phylogenetic tree analysis showed the FnERF017protein was highly homologous to Fragaria vesca ERF protein. Colletotrichum gloeosporiodes stress could induce up-regulation of FnERF017, Which indicated that it was in positive response to C. gloeosporiodes stress. The prediction of promoter cis acting elements showed that the promoter sequence pFnERF107 contained many types of cis acting elements, such as hormone response elements, and stress response elements. It provids a basis for further research on the function of Fn ERF017 gene.
