Abstract:
In this study, gastric cancer SGC-7901 cells were treated with Hedyotis diffusa injection in different concentrations in order to detect the effect on cell proliferation. Trypan blue and giemsa staining method were used to detect the effects of different concentrations of Hedyotis diffusa on SGC-7901 cell viability and cell clone formation. The cell proliferation rate of SGC-7901 after drug treatment was detected by EdU method.RT-qPCR and Western blot were also performed to determine the mRNA and protein levels of CCNB1 and CCND1. The results showed that Hedyotis difflosa injection could inhibit the cell viability of SGC-7901, and the cell viability decreased with the increase of drug concentration. Human gastric cancer cells SGC-7901 were treated with different concentrations of Hedyotis diffusura injection for 48 h, and the cell clone formation rate and cell proliferation rate were sig-nificantly decreased. RT-qPCR and Western blot analysis showed that the mRNA and protein levels of CCNB1 and CCND1 were significantly downregulated by Hedyotis diffusa injection in different concentrations. With the higher drug concentration, the expressions of CCNB1 and CCND1 showed a declining trend. In conclusion, Hedyotis diffusa suppresses the proliferation of gastric cancer SGC-7901 cells by inhibiting the expression of CCNB1 and CCND1.
