Abstract:
To investigate the effect of dandelion flavonoids on antioxidant enzyme activity and gene expression in mice, fifty 5-week-old SPF ICR male mice(22 g±2 g) were divided into a while the control group, control group, high concentration of dandelion flavonoid group(100 mg ·kg

-1), medium concentration of dandelion flavonoid group(50 mg ·kg
-1 ·d
-1), low concentration of dandelion flavonoid group(25 mg ·kg
-1 ·d
-1), test group was given different concentrations of dandelion flavonoid once a day by regular gavage, while the control group was given the same amount of normal saline for 4 weeks, control group was given the same amount of normal DMSO for 4 weeks.The activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX), total antioxidant capacity(T-AOC), and malondialdehyde(MDA) content were determined in the serum and liver tissues of mice, and the relative mRNA expression of SOD-1, SOD-2, GPX-1, GPX-4 in liver tissues of mice were determined by RT-qPCR. The results show that dandelion flavonoids can significantly increase the activities of SOD, GSH-PX, T-AOC in mouse serum and liver tissue increased significantly(P<0.05), the content of MDA reduced significantly(P<0.05), and the expression of SOD-1, SOD-2, GPX-1, GPX-4 mRNA increased significantly. Research shows, dandelion flavonoids can improve the antioxidant capacity of mice and have anti-aging effects.
