2019年广西鸽Ⅰ型副黏病毒的分离鉴定及F基因分子进化分析

Isolation,Identification and Molecular Evolution Analysis of F Gene of Pigeon Paramyxovirus Type 1 Isolated From Guangxi in 2019

  • 摘要: 为了解广西鸽Ⅰ型副黏病毒(pigeon paramyxovirus type 1,PPMV-1)分离株的基因亚型及遗传进化,2019年从广西鸽场疑似感染PPMV-1的送检临床样品进行病毒分离和鉴定,并对其进行F基因的测序以及系统发育和分子钟的分析。结果显示,从送检样品分离3株病毒株,分别命名为Pi/CH/GX1101/2019,Pi/CH/GX1201/2019和Pi/CH/GX1202/2019。测序结果表明其F基因编码区长度均为1 662核苷酸(nt),编码553氨基酸(aa)。分离株的F蛋白裂解位点为112RRQKRF117,符合强毒株特征;系统发育分析表明3株毒株属于两个基因亚型:Ⅵ.2.1.1.2.1亚型(原Ⅵj)和Ⅵ.2.1.1.2.2亚型(原Ⅵk);分子钟分析结果显示中国Ⅵ.2.1.1.2.1亚型和Ⅵ.2.1.1.2.2亚型毒株大约在20世纪80年代开始出现进化分歧;Ⅵ.2.1.1.2.1亚型的Pi/CH/GX1201/2019和Pi/CH/GX1202/2019与位于进化枝低节点的野鸟源参考毒株W4的亲缘关系较近,同在一个分支中,而Ⅵ.2.1.1.2.2亚型的Pi/CH/GX1101/2019毒株与进化枝低节点的2株野鸟源参考毒株Grey heron/CH/GD/GZ333/2015和European turtle dove/CH/GD/GZ23/2015的亲缘关系相近,同在一个小簇中。研究结果证明广西鸽群主要流行Ⅵ.2.1.1.2.1亚型和Ⅵ.2.1.1.2.2亚型的PPMV-1,并推测存在从野鸟传入商业鸽群的风险。

     

    Abstract: In order to understand the sub-genotype and genetic variation of pigeon paramyxovirus type 1(PPMV-1) isolates in Guangxi, three strains of the virus were isolated from clinical samples suspected to be infected with PPMV-1 in Guangxi pigeon farms in 2019. The isolates were subjected to F gene sequencing, phylogenetic analysis and molecular clock phylogeny analysis. The results showed that 3 strains were isolated and identified from clinical samples and named Pi/CH/GX1101/2019, Pi/CH/GX1201/2019 and Pi/CH/GX1202/2019, respectively. Sequencing analysis of the isolates showed that F gene ORF length of strains was 1 662 nt coded 553 amino acids and the cleavage site of F protein was 112RRQKRF117, which possesses the virulent NDV molecular characteristics. The three isolates identified in this study were grouped into genotype Ⅵ. Phylogenetic analysis showed that the three strains belonged to two genotypes: sub-genotype Ⅵ.2.1.1.2.1(former Ⅵj genotype) and sub-genotype Ⅵ.2.1.1.2.2(former Ⅵk genotype). The molecular clock phylogeny analysis showed that the Chinese sub-genotype Ⅵ.2.1.1.2.1 and Ⅵ.2.1.1.2.2 strains began to diverge evolutionarily around the 1980 s. Pi/CH/GX1201/2019 and Pi/CH/GX1202/2019(sub-genotype Ⅵ.2.1.1.2.1) were found to be more closely related to the low-nodal wild-bird-origin strain W4 in the same branch. The Pi/CH/GX1101/2019 strain(sub-genotype Ⅵ.2.1.1.2.2) was also found to be related to 2 wild-bird-origin strains(Grey heron/CH/GD/GZ333/2015 and European turtle dove/CH/GD/GZ23/2015) at the low node and in the same small cluster. The results of the study showed that Ⅵ.2.1.1.2.2 and Ⅵ.2.1.1.2.1 were the predominant sub-genotype circulating in pigeons in Guangxi and there was a presumed risk of wild bird isolates entering commercial pigeon populations.
