威氏绿绒蒿(Meconopsis wilsonii)花色相关基因MwF3H的克隆及表达分析

Cloning and Expression Analysis of Flower Color Related Gene MwF3H from Meconopsis wilsonii

  • 摘要: 通过cDNA末端快速扩增技术(rapid amplification of cDNA ends, RACE),从威氏绿绒蒿(Meconopsis wilsonii)花中克隆得到黄烷酮-3-羟化酶(flavanone-3-hydroxylase, F3H)基因,将其命名为MwF3H,对其理化性质、蛋白结构域、进化关系、基因表达水平进行分析,并探讨MwF3H基因在威氏绿绒蒿花发育不同时期的表达模式。结果表明,该基因编码区长度为1 104 bp,其编码蛋白的分子量为41.444 kDa, 367个氨基酸,理论等电点(pI)4.98,属于PLNO2515超基因家族,构建系统发育树发现威氏绿绒蒿与藏南绿绒蒿(M.zangnanensis)亲缘关系较近,同一物种不同地理区域存在差异。qPCR结果显示MwF3H基因在威氏绿绒蒿花发育不同阶段均有不同程度的表达,除花蕾着色期外,盛花期表达量最高。研究表明MwF3H基因可能参与威氏绿绒蒿花相关色素的生物合成,在威氏绿绒蒿花色形成过程中具有关键作用。

     

    Abstract: The flavanone-3-hydroxylase(F3 H) gene was cloned from the flower of Meconopsis wilsonii by rapid amplification of cDNA ends(RACE) and named MwF3 H. Its physical and chemical properties, protein domain, evolutionary relationship and gene expression level were analyzed to explore the expression pattern of MwF3 H gene in different flowering stages of M. wilsonii. The results showed that the length of the coding region of the gene was 1 104 bp, encoding 367 amino acids, the molecular weight of the protein is 41.444 kDa, and the theoretical isoelectric point(pI) is 4.98, belonging to the PLNO2515 supergene family. The construction of a phylogenetic tree found that M. wilsonii and M. zangnanensis is closely related, and the same species is different in different geographical areas. The qPCR results showed that the MwF3 H gene is expressed to varying degrees in different stages of the development of M. wilsonii, except for the bud coloring stage, the highest expression level is in the full bloom stage. The results showed that MwF3 H gene most probably was involved in the biosynthesis of related pigments during M. wilsonii flowering. It may play a key role in the process of flower color formation.
