染色体外环状DNA的研究进展

Research Progress of Extrachromosomal Circular DNA

  • 摘要: 染色体外环状DNA (extrachromosomal circular DNA, eccDNA)是一类位于染色体外的DNA分子,普遍存在于真核细胞中,并发挥其调控作用。研究表明,eccDNA是从染色体分离或脱落而形成的,独立存在,并可以转录出编码和非编码RNA,广泛参与细胞的生理和病理调控。大量证据显示,在肿瘤的发生发展中,高水平表达的促癌基因来自eccDNA转录。本论文对eccDNA类型、形成机制、eccDNA生理及病理调控机制几个方面进行综述,以促进eccDNA在细胞生理及病理功能和应用方面的研究。

     

    Abstract: Extrachromosomal circular DNA(eccDNA), a kind of DNA molecules located the extrachromosome, widely exists in eukaryotic cells, and in which it plays a regulatory role.Studies have shown that eccDNA formed through chromosome separation or abscission, existing independently, can transcribe coding and non-coding RNAs, which is widely involved in physiological and pathological regulation of cells.Lots of works shows that the high-level expression of oncogenes comes from eccDNA transcription in the occurrence and development of tumors. In order to promote the research of eccDNA in cellular physiological and pathological function and application, this paper reviews the type, formation mechanism, physiological and pathological regulation mechanism of eccDNA.
