Abstract:
In this study, 196 sugarcane pentatricopeptide repeat protein family genes(ScPPR)were identified in the sugarcane(Saccharum spp.) genome based on the transcriptome data of sugarcane mosaic virus(SCMV) infected healthy young leaves, and the basic physicochemical information, subcellular localization, motifs, phylogenetic tree, transcriptome expression of the family members and RT-qPCR experimental verification were analyzed. The results showed that we have identified 196 ScPPR family members in sugarcane according to the transcriptome data, and the molecular weight of ScPPR-encoded proteins ranged from 5 897 to 144 174 kDa, for 52 to 1 293 amino acid residues, and the isoelectric point of these proteins ranged from 4.70 to 10.54. Prediction of the subcellular localization of ScPPR family members showed that most members were localized in chloroplasts and nuclei. Phylogenetic relationship analysis showed that the ScPPR gene family belongs to Group Ⅰ and Group Ⅱ. Gene expression of sugarcane transcriptome showed that the expression of ScPPR2, ScPPR48, ScPPR54, ScPPR58, ScPPR151, ScPPR165, ScPPR168, ScPPR184 were significantly variable in expression. It was also verified by RT-qPCR experiment that ScPPR2, ScPPR54, ScPPR135, and ScPPR165 were highly expressed in sugarcane leaves infected by SCMV. ScPPR23, ScPPR48, and ScPPR57 expression was significantly down-regulated. All in all, the genes we have identified above that play important biological roles in sugarcane resistance to sugarcane mosaic virus infestation. The results have some references significance for the subsequent study of PPR family in sugarcane and the individual study of PPR genes.
