紫九牛总蒽醌对胃癌细胞SGC7901增殖、凋亡、迁移和侵袭作用的影响及其机制研究

Study on the Effect and Mechanism of Total Anthraquinone of Zijiuniu on Proliferation, Apoptosis, Migration and Invasion of Gastric Cancer SGC7901 Cells

  • 摘要: 本研究的目的是探讨紫九牛总蒽醌对人(Homo sapiens)胃癌细胞SGC7901增殖、凋亡、迁移和侵袭作用的影响及其与PI3K/AKT/mTOR通路的相关性。用不同浓度的紫九牛总蒽醌分别处理细胞48 h和72 h后,以CCK-8法检测细胞增殖能力;Hoechst细胞染色与Annexin V-FITC/PI染色检测细胞凋亡情况;划痕实验、 Transwell实验检测细胞迁移与侵袭能力;Western blot检测线粒体途径凋亡蛋白Cleaved-Caspase-3、 BAX、 BCL-2,迁移与侵袭相关蛋白MMP-2、 MMP-9,以及PI3K/AKT/mTOR信号通路相关蛋白的表达。结果显示,与对照组比较,紫九牛总蒽醌对SGC7901细胞增殖有显著抑制作用(P<0.01),抑制率与药物浓度、作用时间呈正相关,48 h与72 h的半数抑制浓度(half inhibitory concentration,IC50)分别为190、 120μg/mL。100、 50μg/mL紫九牛总蒽醌浓度能诱导SGC7901细胞凋亡(P<0.05)。100、 50、 25μg/mL紫九牛总蒽醌能抑制SGC7901细胞迁移能力(P<0.01或P<0.05)。100、 50μg/mL紫九牛总蒽醌能抑制SGC7901细胞侵袭能力(P<0.01或P<0.05)。100μg/mL紫九牛总蒽醌能下调MMP-2、 MMP-9蛋白的表达水平(P<0.05); 100、 50μg/mL紫九牛总蒽醌均能上调Cleaved-Caspase-3、 BAX蛋白的表达水平,下调p-AKT蛋白的表达水平(P<0.01或P<0.05); 100、 50、 25μg/mL紫九牛总蒽醌均能下调p-mTOR、 BCL-2蛋白的表达水平(P<0.01或P<0.05)。本研究发现紫九牛总蒽醌能抑制SGC7901细胞增殖、诱导凋亡并抑制迁移与侵袭,其作用机制与抑制PI3K/AKT/mTOR信号通路有关。

     

    Abstract: The puropse of this study was to investigate the effects of total anthraquinone of Zijiuniu on proliferation, apoptosis, migration and invasion of human(Homo sapiens) gastric cancer SGC7901 cells and its correlation with PI3K/AKT/mTOR pathway. After treating the cells with different concentrations of total anthraquinone of Zijiuniu for 48 h and 72 h, proliferation capacity of SGC7901 cells were detected by CCK-8 method. Apoptosis was detected by Hoechst cell staining and Annexin V-FITC/PI staining. Migration and invasion were detected by scratch test and Transwell test. Apoptosis related proteins Cleaved-Caspase-3, BAX, BCL-2 in the mitochondrial pathway, migration and invasion related proteins MMP-2, MMP-9 and PI3K/AKT/mTOR signaling pathway associated proteins expression were detected by Western blot. The results showed that compared with the control group, the total anthraquinone of Zijiuniu had a significant inhibitory effect on the proliferation of SGC7901 cells(P<0.01), the inhibition rate was positively correlated with drug concentration and action time, the half inhibitory concentration(IC50) of 48 h and 72 h were 190 and 120 μg/mL respectively. The total anthraquinone of Zijiuniu on centration at 100 and 50 μg/mL can induce apoptosis in SGC7901 cells. The total anthraquinone of Zijiuniu on centration at 100, 50 and 25 μg/mL can inhibit the migratory ability of SGC7901 cells(P<0.01 or P<0.05).The total anthraquinone of Zijiuniu on centration at 100 and 50 μg/mL can inhibit the invasion ability of SGC7901 cells(P<0.01 or P<0.05). The total anthraquinone of Zijiuniu at 100 μg/mL can down regulate MMP-2 and MMP-9 proteins expression level(P<0.05). The total anthraquinone of Zijiuniu at 100 and 50 μg/mL can up regulate the expression level of Cleaved-Caspase-3 and BAX protein, and down regulate the expression level of p-AKT protein(P<0.01 or P<0.05). The total anthraquinone of Zijiuniu at 100, 50 and 25 μg/mL can down regulate the expression level of p-mTOR and BCL-2 protein(P<0.01 or P<0.05). The present study found that the total anthraquinone of Zijiuniu could inhibit the proliferation of SGC7901 cells, induce the apoptosis, inhibit the migration and invasion, and its effects were related to the inhibition of PI3K/AKT/mTOR signaling pathway.
