杜仲EuERD16基因的克隆及功能分析

Cloning and Function Analysis of EuERD16 Gene in Eucommia ulmoides

  • 摘要: 脱水诱导早期应答(Early Responsive to Dehydration, ERD)基因是在干旱胁迫下可以快速应答的基因。本研究克隆了杜仲(Eucommia ulmoides)EuERD16基因,对其进行生物信息学分析,采用实时定量PCR技术分析EuERD16基因在杜仲不同组织和胁迫下的表达情况,进而构建过表达载体,采用农杆菌介导法获得过表达EuERD16基因烟草(Nicotiana tabacum)植株,分析EuERD16基因在烟草中的抗旱功能。结果发现,EuERD16基因的CDS序列长度为387 bp,编码128个氨基酸,与拟南芥(Arabidopsis thaliana)AtERD16序列相似性较高,均为1个泛素单体(76个氨基酸)融合1个60S核糖体亚基L40(52个氨基酸),属于泛素延伸蛋白亚族,该基因启动子区域含有光、干旱和低温等响应元件。在杜仲幼苗中,EuERD16基因在茎和根中表达较高,在成年期杜仲树中,EuERD16基因在3月、5月的雌雄叶和7月的雌果中表达较高,并受到20%PEG6000模拟干旱、蓝光和低温胁迫的信号诱导。干旱胁迫下,转EuERD16基因烟草比野生型烟草的萎蔫程度轻,且其CAT、SOD、POD酶活性和脯氨酸含量显著高于对照组的(P<0.05),H2O2、O2-和MDA含量显著低于对照组的(P<0.05)。结果表明EuERD16参与杜仲干旱胁迫反应,为进一步研究杜仲抗旱分子机制提供了参考。

     

    Abstract: Early Response to Dehydration(ERD) gene is a kind of gene that can quickly respond to drought stress.In this study,the EuERD16 gene of Eucommia ulmoides was cloned,and its bioinformatics analysis was carried out.The expression of EuERD16 gene in different tissues and under stress conditions of Eucommia ulmoides was analyzed by real time quantitative PCR technology,and then the over expression vector was constructed.Overexpressed EuERD16 gene tobacco(Nicotiana tabacum) plants were obtained by Agrobacterium-mediated method,and the drought resistance function of EuERD16 gene in Nicotiana tabacum was analyzed.The results showed that the CDS sequence of the EuERD16 gene was 387 bp,encoding 128 amino acids,and had high homology with the AtERD16 sequence of Arabidopsis thaliana.Both of them were one ubiquitin monomer(76 amino acids) fused with one 60S ribosomal subunit L40(52 amino acids),belonging to the ubiquitin extention protein subfamily.The promoter region of this gene contained response elements such as light,drought and low temperature.In Eucommia ulmoides seedlings,EuERD16 gene is highly expressed in stems and roots.In mature E.ulmoides trees,EuERD16 gene is highly expressed in male and female leaves in March and May,and female fruits in July,and is induced by signals of 20% PEG6000 to simulate drought,blue light and low temperature stress.Under drought stress,the wilting degree of EuERD16 transgenic tobacco was less than that of wild type tobacco,and the enzyme activities of CAT,SOD,POD and the content of proline were significantly higher than those of control group(P<0.05),while the contents of H2O2,O2- and MDA were significantly lower than those of control group(P<0.05).The result showed that EuERD16 participated in the drought stress response of E.ulmoides,providing a reference for further study on drought resistance mechanism of E.ulmoides.
