Abstract:
Migrasomeis organelle produced by migrating cells, which play a signal transduction role in embryonic development, immune response, tumor metastasis and other cellular biological behaviors. However, its role in human(Homo sapiens) liver cancer is still unclear. In order to explore the effect of migration related genes on the malignant progression and clinical prognosis of patients with liver cancer, we used TCGA database to analyze the mRNA expression levels of migration related marker genes TSPAN4, TSPAN7, KIF5B, MYO19, DNM1L. Kaplan Meier curve was drawn to analyze the influence of these genes on the clinical prognosis of patients with liver cancer. The gene function enrichment analysis and pathway enrichment analysis were conducted on the website of assistant for clinical bioinformatics(ACLBI). The STRING database platform and Cytoscape software were used to analyze the network of protein-protein interaction(PPI). The siRNA was transfected into human hepatoma cells SMMC7721 and Huh7, and the changes of cell proliferation after transfection with TSPAN4 were evaluated by CCK8 method. The expression and distribution of TSPAN4 protein were detected by immunofluorescence staining; The cell cycle distribution was detected by propidium iodide(PI) single staining. The results showed that the expression of TSPAN4, TSPAN7, KIF5B, MYO19, DNM1L mRNA in hepatocellular carcinoma and normal tissues were significantly different(FDR<0.001), among which the expression of TSPAN4, MYO19, DNM1L mRNA significantly affected the prognosis of patients with liver cancer(P<0.05). The expression of TSPAN4, TSPAN7, KIF5B, MYO19, DNM1L mRNA affects the expression level of multiple genes in patients with liver cancer. Among them, TSPAN4, the most important marker gene of the migrasome, is related to multiple immune related cell proliferation, lipid metabolism, amino acid metabolism and other functions(FDR<0.05), and participates in phagosome, cell cycle, complement system and other signal pathways(FDR<0.05). Knockdown of TSPAN4 inhibited the proliferation of human hepatoma cells SMMC7721 and Huh7, and increased the percentage of cells in G2/M phase. This study demonstrated that the migration related marker genes were related to the prognosis of patients with liver cancer, and TSPAN4 gene affected the proliferation, migration and cell cycle of liver cancer cells.
