Abstract:
Solid-substrate fermentation and enzymolysis of soybean stalk cellulose pretreated with ammonium had been studied.The major affecting factors of solid-substrate fermentation with the cellulase produced by Trichoderma koningii are temperature,pH value,medium moisture content and time.The major factors affecting enzymolysis are pH value,temperature and time.It showed that the adequate condition for enzyme production is temperature 30℃,pH value 5.0,liquid to solid ratio 2.5:1,time 96 h,and cellulase activity produced is 798.84 FPU/g.Using this cellulase for enzymolysis,the optimum condition is:temperature 55℃,pH value 5.6,36 h,the yield of enzymolysis is 18.98%.The composition of enzymolysis product was analyzed by liquid chromatography,which was composed of glucose,xylose and cellobiose.This study provides reference data for further comprehensive use of soybean stalk.
