枯草芽孢杆菌蛋白酶结构与表达载体分析

Analysis of Structure and Expression Vector of Protease from Bacillus Subtilis

  • 摘要: 文章对枯草芽孢杆菌蛋白酶同源建模的三维结构进行分析,并对基因密码子和不同生物细胞载体基因密码子偏好性进行比较,确定枯草芽孢杆菌蛋白酶比较一致的表达载体。结果表明,枯草芽孢杆菌蛋白酶三维预测模型超过90%氨基酸残基落在最适区,属于高质量的预测模型,其主要结构是α-螺旋和不规则结构。对枯草芽孢杆菌蛋白酶基因与不同候选生物细胞载体基因组进行比较,发现巴斯德毕赤酵母是枯草芽孢杆菌蛋白酶最适合的表达载体。

     

    Abstract: This article analyzes the three-dimensional structure of Bacillus subtilis protease homology modeling, and compares gene codons with different biological cell vector gene codon preferences, and determines the more consistent expression vector of subtilis protease. The results show that more than 90% of the amino acid residues of the subtilis protease three-dimensional prediction model fall in the optimal region, which is a high-quality prediction model, and its main structure is α-helix and irregular structure. Comparing the Bacillus subtilis protease gene with the cell vector genomes of different candidate organisms, it is found that Pichia pastoris is the most suitable expression vector for the subtilis protease.
