山西不同产地野生葛根外植体灭菌条件优化分析

Optimum Sterilization Conditions for Radix Puerariae Explants in Different Regions of Shanxi

  • 摘要: 文章对不同产地的野葛根外植体的消毒方法进行了优化。结果表明:外植体材质脆、嫩、表皮浅褐色的大同灵丘野葛根的最佳消毒条件为:质量分数75%乙醇30 s,质量分数0.2%HgCl2 8 min,污染率为8.3%,死亡率为5%;坚实、较嫩、表皮浅褐色的运城平陆野葛的最佳灭菌条件为:质量分数75%乙醇30 s,质量分数0.2%HgCl2 8 min,污染率10.2%,死亡率0;坚实、较硬、表皮粗糙、深褐色的晋城泽州产野葛的最佳灭菌条件为:质量分数75%乙醇30 s,质量分数0.2%HgCl2 10 min污染率5.7%,死亡率32.2%。

     

    Abstract: In this paper, the disinfection method of Radix Puerariae explants in different origin is optimized. The results showed that the optimal disinfection conditions of crisp, tender and light brown skin form in Lingqiu County are 75% ethanol of 30 s, 0.2% HgCl2 8 min, pollution rate of 8.3% and mortality rate of 5%. The best sterilization conditions for the solid, tender and light brown skin form in Pinglu County are 75% ethanol of 30 s, 0.2% HgCl2 8 min, pollution rate is 10.2% and mortality is 0. The best sterilization conditions for wild Radix Puerariae in solid, hard, rough epidermis and dark brown in Zezhou County are 75% ethanol of 30 s, 0.2%HgCl2 10 min pollution rate of 5.7% and mortality rate of 32.2%.
