基于ISSR分子标记的白术遗传多样性分析

Genetic Diversity Analysis of Atractylodes macrocephala Based on ISSR Markers

  • 摘要: 基于ISSR分子标记技术研究17份不同产地白术种质的遗传多样性和亲缘关系。结果表明,筛选出9条多态性引物,平均多态性条带百分率为72%,各种质间遗传相似系数(GS)值在0.589~0.904之间,遗传距离(GD)值在0.084~0.425之间,聚类分析将17份种质划分为7组,其中B 3(湖南省平江县-2)和B 17(浙术1号)材料间亲缘关系最近,B 5(湖北省咸丰县小村乡)和B 14(浙江省新昌县回山乡)材料间亲缘关系最远。17份白术种质资源具有较高的遗传多样性,遗传信息丰富。

     

    Abstract: Genetic diversity and genetic relationships of 17 A. macrocephala germplasms from different place were studied by ISSR markers.The results showed that 9 polymorphic primers were selected, and the average percentage of polymorphic bands was 72%.The genetic similarity coefficient(GS) and genetic distance(GD) of various germplasms ranged from 0.589 to 0.904 and 0.084 to 0.425,respectively.Further, the 17 germplasms were divided into 7 groups by cluster analysis.Among them, B 3(namely Pingjiang-2 from Hunan Povince) and B 17(namely Zheshu 1) had the closest genetic relationship, while B 5(from Xiaocun village, Xianfeng County, Hubei Province) and B 14(from Huishan village, Xinchang County, Zhejiang Province) had the farthest genetic relationship.In conclusion, the 17 A. macrocephala germplasms had high genetic diversity and rich genetic information.
