不同高渗物质对参薯组培苗限制生长的影响

Effects of Different Hypertonic Substances on Growth Restriction of Tissue Culture Seedlings of Dioscorea alata

  • 摘要: 采用单因素试验研究参薯在3种高渗物质(蔗糖、甘露醇、山梨醇)处理下的存活率及生长状态。结果表明,培养150 d时,10 g·L-1蔗糖处理的植株平均高度为(3.55±0.17)cm,显著低于对照及其他处理;30 g·L-1和40 g·L-1甘露醇浓度处理的植株平均高度分别为(3.88±0.19)cm、(2.13±0.19)cm,显著低于对照,综合植株高度、存活率及生长状态,30 g·L-1处理的试管苗明显优于40 g·L-1处理的试管苗;0~40 g·L-1范围内随着山梨醇浓度的增加,与对照相比株高和存活率均无明显差异。10 g·L-1蔗糖处理和30 g·L-1甘露醇处理均可在150 d内有效抑制参薯组培苗的生长。

     

    Abstract: In this study, the single factor experiment was carried out to study the survival rate and growth status of Dioscorea alata.treated with 3 kinds of hypertonic substances, and the results showed that at 150 days of culture, the average height of plants treated with 10 g·L-1 sucrose was(3.55±0.17) cm, which was significantly lower than that of the control and other treatments. In addition, the average height of plants treated with 30 g·L-1 and 40 g·L-1 mannitol was(3.88±0.19) cm and(2.13±0.19) cm, respectively, which were significantly lower than that of the control.Comprehensively considering the height, survival rate and growth status of plants, the treatment with 30 g·L-1 was significantly better than that treated with 40 g·L-1. Also, in the range of 0-40 g·L-1,there was no significant difference in plant height and survival rate compared with the control with the increase of sorbitol concentration. Both 10 g·L-1 sucrose and 30 g·L-1 mannitol treatment could effectively inhibit the growth of tissue culture seedlings of D. alata within 150 days.
