Abstract:
Taking the Auricularia cornea var. Li strain provided by the Guizhou Province Edible Fungus Engineering Technology Research Center as the research object, DNA sequencing was performed to determine the reliability of its provenance, and the optimal nutrient conditions for its mycelial growth were screened. It hopes to provide a basic theoretical basis for the introduction and demonstration of Auricularia cornea var. Li in Guizhou. The identification shows that the similarity between the tested strain and Auricularia polytricha was more than 99%, so it is a branch under Auricularia polytricha. Further research shows that the growth conditions of Auricularia cornea var. Li hyphae are not harsh. It can grow well at 25-35 ℃, among which the optimum temperature is 30 ℃, which is a medium-high temperature edible fungus. Its suitable carbon sources are soluble starch, sucrose and maltose, and soluble starch is the best. Its suitable nitrogen sources are soybean meal powder, peptone and yeast extract, and soybean meal powder is the best. In this paper, two suitable carbon and nitrogen sources were selected, and a 4-factor 3-level orthogonal experiment was carried out. In the end, the best combination of carbon and nitrogen source concentration for the growth of Auricularia cornea var. Li mycelium is 10.0 g soluble starch, 10 g sucrose, 2.5 g soybean meal powder, and 1.5 g peptone.
